明胶酶A、B蛋白在不同大小腹主动脉瘤组织中表达的实验研究

摘　　要：目的 :探测明胶酶A (MMP - 2 )和明胶酶B(MMP - 9)的蛋白在不同大小腹主动脉瘤 (AAA)组织中表达高低的情况 ,证实在不同大小AAA组织中两者的蛋白表达高低不同。方法 :用MMP - 2和MMP - 9的单克隆抗体 ,以免疫组织化学SABC方法在实验组 2 6例不同横径大小的AAA组织 (7例小AAA :4 0～ 5 5cm ,11例中等大小AAA :5 6～ 6 5cm ,8例大AAA :>6 5cm)和对照组 10例正常腹主动脉组织的切片上探测MMP - 2和MMP- 9抗原 (蛋白 )。结果 :MMP - 2蛋白的表达在小AAA和中AAA高于对照组 (P <0 0 1) ,大AAA与对照组无显著性差异 (P >0 0 5 ) ,小AAA高于中AAA(P <0 0 1) ;MMP - 9的表达小、中、大AAA组织中均高于对照组 (P <0 0 1) ,中AAA的表达高于小和大AAA(P <0 0 1) ,小AAA和大AAA无显著性差异 (P >0 0 5 )。结论 :在横径≤6 5cm的AAA组织中 ,MMP - 2和MMP - 9两者的蛋白共同过度表达 ,在横径 >6 5cm的AAA组织中 ,MMP -9过度表达。在横径 5 6～ 6 5cmAAA组织中 ,MMP 9蛋白的表达高于横径 4 0～ 5 5cm和横径 >6 5cm的AAA组织。OBJECTIVE:To explore the expression of gelatinase A and B proteins in human abdominal aortic aneurysm (AAA) tissues of various transverse diameters,and to confirm in various size AAA the different expression level. METHOD:Immunohistochemistry was applied to detect protein expression of gelatinase A(MMP-2) and B (MMP-9)in the tissue sections of 26 cases of AAA tissues of various transverse diameters (7 small AAA 4 0～5.5 cm,11 medial AAA 5.6～6.5 cm, and 8 large AAA>6.5 cm in the test group) and 10 cases of normal human abdominal aorta in the control group.RESULTS:The expression of MMP-2 protein in small AAA and middle AAA is significantly higher than in normal abdominal aorta tissues (P<0 01), and there is no signgficant difference between large AAA and normal abdominal aorta tissues (P=1 00>0 05). In small AAA is significantly higher than in medial AAA (P<0 01).The expression of MMP-9 in small,middle and large AAA is significantly higher than that in the control group (P<0.01).The expressim of it in middle AAA is higher than in small and large AAA (P<0 01),and there is no difference in small AAA compare with large AAA (P=0 330>0 05).CONCLUSION:In AAA tissues of transverse diameters less than and equal to 6.5cm MMP-2 and MMP-9 overexpress jointly.In AAA tissues of transverse diameters more than 6.5cm,MMP-9 overexpress.In AAA tissues of transverse diameters from 5.6 to 6.5 cm,the expression of MMP-9 proteins is higher than that in AAA tissues of transverse diameters from 4.0 to 5.5cm and more than 6.5cm.
