摘 要:目的:寻找新的TIL刺激物,摸索迅速解除TIL抑制状态和提高扩增效率的最佳条件。方法:通过黄芪、干地黄水提物联合细胞因子,采用3H-TdR掺入法及流式细胞仪对9例食管癌TIL的增殖及表型进行了分析。结果:在促增殖作用方面,黄芪联合IL-2、IL-2+IL-4、IL-2+TNF-a均较单用其细胞因子有明显增强作用。而干地黄对TIL增殖无明显协同作用。食管癌TIL表型分析发现CD4+T细胞为主,诱导后CD4+T细胞明显增多,CD4/CD8比值由2.8:1上升至9.9:1。结论:黄芪有明显协同细胞因子促进食管癌TIL增殖的作用,激活的食管癌TILCD4+T细胞占绝对优势。Objective: To find out new activator of tumor infiltrating lymphocytes from esophageal cancer, to relieve the repression of TILs and to explore the best condition of TIL proliferation. Methods: TILs were isolated from 9 patients with esophageal cance. With water extract of Astragalus Membranaceus(Aw) or Rehamnnia Glutinosa (RG) combining cytokines the proliferation and phenotypes ot the TILs were investigated by [3H] thymidine incorporation method and flow cytometry. Result: The proliferation of TILs under AM combining with IL-2、IL-2+IL-4、IL-2+TNF-a were significantly stronger than those of the cytokines alone(P<0.05 or P<0.01). However RG combining cytokines showed no effect. It was found that the phenotypes of the TILs were mainly composed of CD4+T cell. the activation CD4+T cell evidendy increased, and the ratio of CD4/CD8 was increased from 2.8:1 to 9.9:1. Conclusion: The results suggested that the AM combining cytokines have the function of promoting proliferation of the TILs. The CD4+T cell is predominant in the astivated TILs.
