摘 要:目的:探讨尿路上皮癌胚抗原Ⅰ(UCA1)mRNA在舌鳞状细胞癌中的表达及其与舌鳞癌病理分级和临床分期的关系。方法:采用SYBR Green II实时荧光定量反转录聚合酶链反应实验方法,从6种基因中筛选出在40对舌鳞癌组织及配对正常舌组织样本中表达有明显差异的基因UCA1,扩大样本量至93例,检测UCA1 mRNA在两者中的表达。应用SPSS13.0软件包分析UCA1 mRNA表达量与舌鳞癌患者临床病理学特征的关系。结果:UCA1 mRNA在93例患者的肿瘤组织中平均表达量为0.7213,在正常舌组织中为0.2756,表达量有显著差异(P<0.001);UCA1 mRNA的表达与患者的性别、年龄、肿瘤大小无关(P>0.05),与舌鳞癌的病理分级和临床分期有显著相关性(P<0.05)。结论:UCA1 mRNA高表达与舌鳞状细胞癌密切相关,对舌鳞癌的诊断、判断病理分级和临床分期具有重要意义。PURPOSE: To investigate the expression of UCA1 mRNA in tongue squamous cell carcinoma (TSCC) and discuss the relationship between UCA1 and the pathological grade and clinical stage of TSCC. METHODS: The SYBR green II real-time fluorescent quantitative RT-PCR was used to screen out UCAI from 6 genes, which expressed markedly higher in tumor tissues than normal tissues of 40 patients with TSCC. Then, the sample size was increased to 93 and the expression of UCA1 mRNA in both tumor and normal tissues were detected. The relationship between expression of UCA1 mRNA and the clinicopathologieal characteristics of TSCC patients was analyzed statistically with SPSS 13.0 software package. RESULTS: Of 93 patients with TSCC, the mean expression of UCA1 mRNA was 0.7213 in tumor tissues and 0.2756 in normal tissues. The difference of UCA1 mRNA expression in tumor and normal tissues was statistically significant (P〈0.001). The expression of UCA1 in TSCC was significantly related to the pathological grade and clinical stage of TSCC (P 〈0.05), but no statistical difference was found in different sex, age and tumor size (P 〉0.05). CONCLUSION: A higher expression of UCA1 mRNA is closely related to TSCC, As a potential tumor molecular marker, UCA1 is of important clinical significance in the diagnosis of TSCC and the judgement of pathological grade and clinical stage.
