摘 要:目的探讨不同病理级别人脑神经胶质瘤组织中p57kip2/p21cip1mRNA的表达变化及其关系。方法采用实时荧光定量PCR检测p57kip2/p21cip1mRNA在68例脑胶质瘤组织和16例非肿瘤脑组织中的表达水平。结果①p57kip2mRNA在脑胶质瘤中的表达水平显著低于非肿瘤脑组织(P<0.01),且其表达水平与肿瘤病理级别呈显著负相关(rs=-0.495;P<0.01)。②p21cip1mRNA在脑胶质瘤中的表达水平与非肿瘤脑组织相比无显著差异(P>0.05),但其表达水平与肿瘤病理级别呈显著负相关(rs=-0.615;P<0.01)。结论p57kip2/p21cip1的异常表达可能与人脑胶质瘤的发生和发展有密切相关,其表达水平可反映肿瘤的恶性程度。ObjectiveTo investigate the changes of p57^kip2/p21^cip1mRNA expressions in gliomas of different WHO histopathological grade and their relationship.MethodsThe glioma tissues were collected from 68 patients with glioma, of whom, 31 patients were astrocytomas(WHO gradeⅡ), 23 were anaplasticastrocytomas(WHO gradeⅢ) and 14 were glioblastomas(WHO gradeⅣ), from June2003 to December 2010. The non-tumor brain tissues were collected from 16 patients with traumatic brain injury or cerebral hemorrhage who underwent microsurgery from June 2003 to January 2010. The mRNA expressions of p57^kip2/p21^cip1were detected by real-time PCR.ResultsThe expression level of p57^kip2in non-tumor brain tissues(100.0%) was significantly higher than that(52.6%) in astrocytoma tissues(P〈0.01) which was significantly higher than that(15.8%) in anaplasticastrocytoma tissues. The expression level of p21^cip1in astrocytoma tissues(84.3%) was significantly higher than that(19.6%) in anaplasticastrocytoma tissues(P〈0.01). There were no significant difference in the expression level of p57^kip2between anaplasticastrocytomas and glioblastomas(P〈0.05). There was also no significant difference in the expression level of p21^cip1between non-tumor brain tissues and astrocytoma tissues(P〈0.05), and between anaplasticastrocytoma and glioblastoma tissues(P〈0.05).Both the expression levels of p57^kip2and p21^cip1were significantly negatively correlated with the grade of gliomas(rs=-0.495 andrs=-0.615, respectively;P〈0.01).ConclusionsThe mRNA expressions of both p57^kip2and p21^cip1are downregulated in gliomas. Their expression levels are negatively correlated with the histopathological grade of glioma.
