摘  要:目的观察人参皂苷Rb1对SweAPP-SK细胞的保护作用。方法用Rb1处理SweAPP-SK细胞,用MTT分析、酶联免疫分析和流式细胞术观察Rb1对SweAPP-SK细胞增殖、Aβ分泌量、细胞活性氧(ROS)水平和Ca2+水平的影响。结果较SK-N-SH细胞,SweAPP-SK细胞增殖明显延迟,细胞ROS及Ca2+水平增加。用Rb1处理SweAPP-SK细胞后,细胞增殖加快,细胞外Aβ40浓度明显降低,细胞ROS及Ca2+水平明显降低。结论 Rb1对SweAPP-SK细胞有保护作用,该保护作用的发挥可能与其能减少Aβ分泌及降低细胞内ROS和Ca2+水平有关。Objective To explore the protective effects of Ginsenoside Rbl on SweAPP-SK cells, which were constructed using human neuroblastoma SK-N-SH cells, had a high expression level of amyloid precursor protein (APP) and secreted more β-amylold peptide ( Aβ). Methods SweAPP-SK cells were treated with Rbl, and MTT assay, ELISA and flow cytometry were used to examine cell proliferation, Aβ secretion, and intracellular reactive oxygen species (ROS) and Ca2+ levels. Results SweAPP-SK cells proliferated slower and had higher levels of ROS and Ca2+ compared with SK-N-SH cells. After treated with Rbl, SweAPP-SK cells showed a faster proliferation, secreted less Aβ, and had lower levels of ROS and Ca2+ in comparison with the untreated. Conclusions Rbl has neuroprotective effects, which maybe associate with its abilities to decrease Aβ secretion by neurocytes and levels of ROS and Ca2+ in them.
