疏肝健脾方药对NASH大鼠肝组织p38MAPK mRNA及蛋白表达的影响

摘　　要：目的观察疏肝健脾方药对非酒精性脂肪性肝炎(NASH)大鼠肝组织p38丝裂原激活蛋白激酶(p38MAPK)mRNA和蛋白表达的影响,探讨疏肝健脾方药抗NASH的作用机制。方法 72只SPF级SD雄性大鼠,适应性喂养1 w,随机分为8组。采用高脂饲料喂养建立NASH大鼠实验模型;造模同时,各用药组分别灌服高、低剂量的疏肝方、健脾方和疏肝健脾合方进行干预。16 w后处死大鼠,检测血脂、肝脂和肝功能;HE和油红O染色观察肝组织病理变化;RT-PCR和Western印迹检测肝组织p38MAPK mRNA和蛋白表达水平。结果与正常组比较,模型组肝小叶结构模糊,脂滴大量沉积,肝细胞肿胀,小叶内、汇管区炎细胞浸润。血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C),肝组织TC、甘油三酯(TG)及p38MAPK mRNA和蛋白表达均显著升高(P<0.05,P<0.01);与模型组比较,用药组大鼠血脂、肝脂、肝组织p38MAPK mRNA和蛋白表达有不同程度降低(P<0.05,P<0.01)。结论疏肝健脾方药能够下调肝组织p38MAPK mRNA和蛋白的表达水平,p38MAPK可能是疏肝健脾治法方药抗NASH的有效作用靶点之一。Objective To discuss the mechanism of soothing liver arid invigorating spleen methods on the expressions of p38 mito- gen-activated protein kinase（p38MAPK）mRNA and protein in rats with non-alcoholic steatohepatitis （NASH）. Methods 72 SPF SD male rats were adaptively fed for 1 w, and divided into 8 groups randomly. The experimental rat modals of NASH were made by fed them high-fat diet. Meanwhile, each treatment group was respectively filled with high and low dose of soothing the liver formula, invigorating spleen formu- la and soothing liver and invigorating spleen formula as intervention. The expressions of blood lipid, hepatic lipid and liver function were de- termined. Also, pathological changes in hepatic tissue were observed by HE and oil red O staining, and real time Q-PCR and Western blot were used to detect the expressions of p38MAPK mRNA and protein respectively in hepatic tissue. Results Compared with that of normal group, hepatic lobule structure of model group was fuzzy, with a lot of lipid droplet deposit, its liver cells were swelling, and inflammatory cells infiltration within the lobule, collect abbacy. The serum and liver levels of TC, LDH-C, the expressions of p38MAPK mRNA and protein were increased significantly （P 〈 0.05, P 〈 0.01 ）. Compared with those of model group, blood lipid, hepatic lipid and the expressions of p38 MAPK mRNA and protein in liver tissue from the treatment group rats were decreased in different degree （P 〈 0.05, P 〈 0.01 ）. Conclusions The expressions of p38MAPK mRNA and protein in the liver could be reduced by soothing liver and invigorating spleen methods and recipes, and p38MAPK might be one of the effective targets of soothing liver and invigorating spleen methods and recipes of NASH.
