摘  要:目的探讨逆向诱导半胱氨酸丰富蛋白(reversion inducing cysteine rich protein with kazal motifs,RECK)基因和基质金属蛋白酶9(matrix metalloproteinase-9,MMP9)在34例外耳道鳞状细胞癌(squamous cell carcinoma,SCC)组织和20例正常外耳道皮肤中的表达情况。方法采用免疫组化SP法和免疫荧光法检测34例外耳道SCC及20例正常外耳道皮肤组织中RECK与MMP9的表达。结果外耳道鳞状细胞癌组织中RECK表达阳性率为44.1%(15/34),正常外耳道皮肤中阳性表达率为80.0%(16/20),两组比较差异有统计学意义(χ2=7.2492,P<0.01);外耳道鳞癌组织中MMP9表达阳性率为67.6%(23/34),正常外耳道皮肤中阳性表达率为25.0%(5/20),两组比较差异有统计学意义(χ2=11.1137,P<0.01)。外耳道鳞状细胞癌组织中RECK和MMP9的表达与组织学分级和肿瘤分期密切相关(P<0.05),而在不同年龄和性别的患者中其表达无差异(P>0.05)。鳞状细胞癌组织分化越差,RECK表达越低,而MMP9表达越高。RECK和MMP9的表达呈负相关(r=-0.453,P<0.05)。结论 RECK基因和MMP9蛋白可能参与了外耳道鳞状细胞癌的发生和发展。OBJECTIVE To investigate the expression of RECK and MMP9 in the carcinogenesis of the external auditory canal squamous cell carcinoma. METHODS Immunofluorescence and immunohistochemical method was used to detect the expression of RECK and MMP9 proteins in 34 external auditory canal squamous cell carcinoma tissues and 20 normal external ear canal skin tissues. RESULTS The positive rate of RECK in the external auditory canal squamous cell carcinoma tissues was 44.1% (15/34) , lower than that in the normal external ear canal skin tissues 80.0% (16/20, P〈 0.05) . Positive rates of MMP9 were 67.6% (23/34) and 25.0% (5/20) respetively (P〈0.05) . The positive rates of RECK and MMP9 were correlated with the histological grade and tumor grade, but no relationship was found with age and sex. The expression of RECK was higher (P〈 0.05) , while MMP9 was lower in the well-differentiated squamous cell carcinoma (P 〉 0.05) . The expression rates of RECK and MMP9 were negatively correlated (r=-0.453, P〈0.05) . CONCLUSION RECK and MMP9 may be involved in the carcinogenesis of the external auditory canal squamous cell carcinoma.
