摘  要:目的探讨色氨酸羟化酶2(THP2)~4570625和~4565946位点单核苷酸多态性在卒中后焦虑(poststrokeanxietydisorde~,PSA)发病机制中的作用。方法通过汉密尔顿焦虑量表(HAMA)将急性缺血性脑卒中患者为PSA组(112人)和对照组(246人),采用聚合酶链反应.限制性片段长度多态性(PCR—RFLP)技术分析两组基因型及等位基因的差异。结果rs4570625G等位基因与PSA的发病增加有关(0R=1.37,P=0.049);女性患者的GG基因型和等位基因均显著高于对照组(p=0.025和P=0.035,OR=1.721,P=0.009)。,两组间rs4565946等位基因和基因型的差异无统计学意义。G.C单倍体的患者PSA的发病危险性增加(OR=1.89,P〈0.01)。结论TPH2基因多态性可能与PSA的发病相关。Objective To investigate the association of two functional polymorphisms of tryptophan hydroxylase 2(THP2) gene, rs4570625 and rs4565946, with poststroke anxiety disorders. Methods Totally 112 poststroke anxiety patients and 246 non-anxious stroke controls were included and completed Hamihon Anxiety Rating Scale. DNA was extracted from blood and genotyped for the two polymorphisms of THP2 gene by polymor- pbism chain reaction. Results The G allele of rs4570625 was associated with the increased risk of poststroke anx- iety. Both the GG genotype and G allele were observed to be significantly higher in female case than in control. No significant difference in genotype mid allele frequencies of the rs4565946 was found among the groups. Patients with the G-C haplotype had significantly increased the risk of poststroke anxiety cored to controls. Conclusions Our findings suggest ttmt these functional polyrnorphisms in TPH2 gene may be involved in development of poststroke arciety.
