摘 要:目的探讨维生素D受体(VDR)在结肠癌上皮细胞中的表达水平对细胞迁移和侵袭能力的影响。方法构建VDR-小干扰RNA(siRNA)干扰质粒,用G418筛选建立结肠癌上皮细胞的VDR-siRNA稳定株,采用Western blot和实时定量PCR方法检测细胞中VDR蛋白和mRNA的表达水平。进一步通过伤口愈合实验和Transwell实验观察VDR表达水平降低对细胞迁移和侵袭的影响。结果 siRNA干扰质粒能显著抑制VDR的表达,获得了VDR-siRNA稳定细胞株,表明VDR表达水平的降低能够促进细胞的迁移能力。结论 VDR能够影响结肠癌细胞的转移能力,为进一步寻找结肠癌基因治疗提供了新的实验依据。Objective To investigate the relationship of vitamin D receptor (VDR) expression with the abilities of cell migration and infiltration in colon epithelial cells. Methods First, the VDR-small interfering RNA (siRNA) interference plasmids was established. Colon epithelial cells VDR- siRNA stable strains were screened and established by G418. Secondly, the Western blot and the realtime PCR methods were adopted respectively to detect the VDR protein in cells and the expressional levels of mRNA. Furthermore, by implementing the experiments of wound healing and Transwell assay, the effects of the decreased expressional level of VDR on the cellular migration and invasion were observed and determined. Results siRNA interference plasmids can significantly inhibit the expression of VDR, and VDR-siRNA stable strains were obtained consequently in this way. The ex- perimental result indicates the reduced expressional level of VDR facilitated the cellular migration obviously. Condusion Our results proved that VDR can influence the metastasis of colon cancer cells, and provided new experimental evidence for further research on gene therapy in colon cancer.
