摘 要:目的:探讨盐霉素对鼻咽癌细胞CNE2的促凋亡作用及其可能的机制。方法:CCK8法检测细胞增殖活性;迁移实验观察迁移能力的改变;比色法检测caspase-3、caspase-8、caspase-9酶活性;流式细胞术检测线粒体膜电位及凋亡率的改变。结果:CCK8结果显示,盐霉素对CNE2细胞的抑制作用呈时间-剂量依赖性关系;迁移实验结果显示盐霉素对CNE-2细胞的迁移抑制作用呈剂量依赖性关系;caspase-3、caspase-8、caspase-9活性随着药物浓度增加而升高,差异具有统计学意义;细胞流式术检测显示,随着盐霉素浓度的增加,线粒体膜电位显著下降,细胞凋亡率增加。结论:盐霉素在体外可以有效抑制鼻咽癌CNE-2细胞增殖并诱导其凋亡。Objective To investigate effect of salinomycin on proliferationl and apoptosis of nasopharyngeal carcinoma CNE-2 cell line. Methods CCK-8 method was used to detect the effect of salinomycin on proliferation. Tumor cell migration was analyzed using transwell chambers provided with an 8um pore polycarbonate membrane. Activation of caspases was measured by the OD values obtained by spectrophotometer at a wavelength of 405 nm. The apoptosis rate and mitochondrial membrane potential (MMP) were analyzed by flow cytometry. Results CCK-8 assay showed exposure to salinomycin resulted in dose- and time-dependent inhibition of CNE-2 cell viability. After exposure to salinomycin for 24 h, a dose-dependent decrease in number of cells migrating through the polycarbonate membrane resulted. Salinomycin with high concentration resulted in increased activity of caspase-3, caspase-8 and caspase-9. Salinomycin induced strongly augmented number of apoptotic cells and reduced the MMP in a dose-dependent compared with the control group. Conclusion Salinomycin can effectively inhibit CNE2 cell proliferation and induce apoptosis.
