摘 要:目的探讨去脂软肝方对高脂诱导脂肪肝大鼠肝组织血管性血友病因子的影响作用。方法高脂饮食复制大鼠脂肪肝模型,将实验鼠随机分为三组:正常组,模型组,去脂软肝方组。去脂软肝方组予去脂软肝方药液灌胃(1 mL/200 g)共12周;HE染色观察肝脏脂肪变性情况;测定大鼠血清血脂、肝脂、血浆血小板聚集率及肝组织血管性血友病因子(vWF)。结果 12周末模型组大鼠血脂[血清总胆固醇(TC)、三酰甘油(TG)]水平比正常组高,差异有统计学意义(P<0.05);模型组大鼠肝组织匀浆(肝脏TC、TG)与比正常组高,差异有统计学意义(P<0.05);去脂软肝方组血清脂质和肝组织脂质水平比模型组低,差异有统计学意义(P<0.05);模型组大鼠血小板聚集率比正常组高,差异有高度统计学意义(P<0.01);模型组大鼠肝脏组织中vWF比正常组高,差异有高度统计学意义(P<0.01);去脂软肝方组血浆血小板聚集率变化及肝组织vWF比模型组低,差异有高度统计学意义(P<0.01)。结论去脂软肝方能明显改善高脂诱导脂肪肝大鼠血脂、肝脂及肝组织脂肪变性。减少vWF分泌、抑制血小板聚集、减轻血管及肝窦内皮细胞损伤可能是其作用机制之一。Objective To investigate the impact of Quzhi Ruan'gan Prescription in von Willebrand factor (vWF) of rats with fatty liver caused by high fat. Methods The model of rats with fatty liver was made by high fat diet, and the model rats were randomized to 3 groups: normal group, model group and Quzhi Ruan'gan Prescription group. Quzhi Ruan'gan Prescription group was treated with garage of Quzhi Ruan'gan Prescription (1 mL/200 g) for 12 weeks. The condition of fatty degeneration of liver was observed with HE staining. The serum lipid, liver lipid, the rate of serum platelet aggre- gation and vWF in liver tissue were tested. Results At the end of 12th weekend, the levels of serum lipid (TC, TG) in model group were higher than those in normal group, the differences were statistically significant (P 〈 0.05); the levels of serum lipid and liver lipid in Quzhi Ruan'gan Prescription group were lower than those in model group (P 〈 0.05); the rate of serum platelet aggregation in the model group was higher than that of normal group, the difference was high- ly statistically significant (P 〈 0.0l); the vWF in liver tissue in the model group was higher than that of normal group, the difference was highly statistically significant (P 〈 0.01); the rate of serum platelet aggregation and vWF in liver tis- sue of Quzhi Ruan'gan Prescription group were lower than those in model group, the differences were highly statistical- ly significant (P 〈 0.01). Conclusion Quzhi Ruan'gan Prescription can improve the conditions of high fat resulting in serum lipid, liver lipid and liver fatty degeneration of rats with fatty liver obviously. One of the mechanism may be as- sociated with reduction of the secretion of vWF, inhibition of platelet aggregation, lightening the damage of blood vessel and liver sinusoidal endothelial cell.
