摘 要:目的明确乙型肝炎病毒(HBV)前C第17位和29位密码子突变体对e抗原合成和分泌的影响。方法用PCR 方法扩增HBV前C/C基因区,克隆后测序及进行序列分析。将HBV前C区T1862/A1899位点突变体亚克隆人表达 质粒 pGEMT中,经体外翻译后,比较野毒株和变异株的表达产物。结果该前 C区突变株在体外仍能合成 HBV e前体 蛋白。结论前 C区 T1862/A1899突变并不能阻断 HBV e抗原前体蛋白合成,e抗原前体可能在分泌过程中受阻。Objective To investigate the effect of T1862 and A1899 codon mutants of hepatitis B virus (HBV) on the synthesis and excretion of the precursor of hepatitis B e antigen. Methods The entire precore and core region were amplified from the sera of a HBeAg -negative patient with fulminant hepatitis B by polymerase chain reaction (PCR). PCR products were cloned into plasmid pUC 18 and sequenced for analysis of precore mutations.Precore and core sequence of T1862/A 1899 variant were also cloned into expression plasmid pGEMT for in vitro tlanscription and translation study on synthesis and excretion of e antigen. Results T1862 /A1899 variant did not interrupt the synthesis of the precursor of e antigen. Conclusion The absence of e antigen in patients with the folminant hepatitis may be explained by precore codon mutation T1862/A1899, which may exercise its effect on the excretion of the precursor of e antigen, other than the synthesis of precursor protein.
