摘  要:目的 研究急性白血病患儿反复应用重组粒细胞集落刺激因子 (rhG CSF)的药代动力学及其与临床的关系。方法 应用酶联免疫吸附测定 (ELISA)方法检测 46例白血病患儿的血清G CSF浓度。结果 获得 2 9例患儿用rhG CSF后的标本 ,其血清G CSF浓度为 (5 335± 5 5 97) μg/L。反复应用rhG CSF的患儿在下次用药前血清G CSF水平恢复正常 ;少数未应用rhG CSF患儿可检测到较高浓度的G CSF。血清G CSF浓度与粒细胞绝对计数值呈负相关 (r=- 0 8746 ,P <0 0 1)。结论 rhG CSF的药代动力学中存在有粒细胞介导的清除机制 。Objective To investigate the pharmacokinetic progerty after repeated administration of recombinant human granulocyte colony stimulating factor(rhG CSF) in children with acute leukemia and its clinical significance.Methods Serum G CSF concentrations were measured in 46 acute leukemic children by enzyme linked immunosorbent assay.Results The serum G CSF concentration was(5 335±5 597)μg/L in 29 children during rhG CSF treatment . In children received repeted treatment of rhG CSF,serum G CSF levels resumed to normal before next administration.Only few cildren(2/8) who didn't receive any rhG CSF treatment had higher rhG CSF concentrations;The serum G CSF concentrations correlated negatively with absolute neutrophil counts(r=-0 8746, P<0 01).Conclusion There is a granulocyte-madiating clearing mechanism in rhG CSF pharmacokinetics, and repeated administration of rhG CSF was safe without accumuting effect.
