透骨草提取物通过Bcl-2和Bax蛋白诱导人乳腺癌细胞株MCF-7凋亡

摘　　要：目的探讨Bax和Bcl-2蛋白在透骨草提取物诱导人乳腺癌细胞株MCF-7细胞凋亡过程中的作用,为透骨草提取物治疗乳腺癌提供实验依据。方法采用吖啶橙染色方法透骨草提取物对MCF-7细胞形态的影响;采用流式细胞术检测透骨草提取物对MCF-7细胞凋亡率的影响;采用蛋白印迹法检测MCF-7细胞Bcl-2和Bax蛋白的表达。结果 30.5μg/ml透骨草提取物可诱导MCF-7细胞胞体皱缩,胞核固缩、呈新月状且边集,出现凋亡小体。30.5μg/ml透骨草提取物处理的MCF-7细胞凋亡率(22.3±1.2)%明显高于对照组(3.2±1.0)%(P<0.05)。30.5μg/ml透骨草提取物可使MCF-7细胞Bcl-2蛋白表达明显低于对照组(P<0.05),而Bax蛋白表达明显高于对照组(P<0.05)。结论透骨草提取物对MCF-7细胞有诱导凋亡作用,其作用机制可能与下调Bcl-2蛋白表达、上调Bax蛋白表达有关。Objective To determine the effects of Phryma leptostachya L＇ Var Asiatica Hara extract on apopto- sis of human breast cancer MCF-7 cells, and tO explore the roles of Bcl-2 and Bax proteins in inducing apoptosis of MCF-7 cells, thus offering the experimental rationales for the treatment of breast cancer. Methods The acridine or- ange staining assay was employed to determine the effects of the extracts on morphology of MCF-7 cells, flow cytome- try was applied to examine the apoptotic rates. Bcl-2 and Bax proteins expressions in MCF-7 cells were detected with Western blotting assay. Results Phryma leptostachya L＇Var Asiatica Hara extract （30.5 μg/ml） led to MCF-7 cell shrinkage and karyopycnosis, as evidenced by the aggregated crescent nuclei to cell edges. Treatment of Phryma lep- tostachya L＇Var Asiatica Hara extract （30.5 μg/ml） yielded a significantly higher apoptotic rate of MCF-7 cells [（22.3±1.2）% vs. （3.2±1.0）%, P〈0.05] and was associated with attenuated Bcl-2 protein expression yet markedly augmented Bax protein expression compared with the control group （both P〈0.05）. Conclusion Phryma leptostachya L＇Var Asi- atica Hara extract induces MCF-7 cell apoptosis possibly via down-regulating Bcl-2 expression and up-regulating Bax protein expression.
