摘　　要：目的研究磷脂酰肌醇3-激酶(phosphatidylinositol 3 kinase,PI3K)和血管内皮生长因子(vascular endothelial growth factor,VEGF)在大肠癌癌变过程中的表达及临床意义。方法选取72例大肠癌患者的大肠癌组织和癌旁正常组织,应用免疫组化法和RT—PCR法检测PI3K和VEGF在大肠癌中的蛋白表达和基因表达水平,并进行统计学分析。结果在大肠癌组织和癌旁正常黏膜组织中PI3K和VEGF的阳性表达率差异均有统计学意义(x^2=60.66,x^2=83.70,P均<0.05),且两者在大肠癌组织中的表达呈正相关(r=0.872,P<0.05)。大肠癌新鲜组织中PIK3R1和VEGFA基因的表达量显著高于癌旁正常黏膜组织中的表达量,且差异均具有统计学意义(t=-2.75,t=-2.61,P均<0.05)。结论 PI3K和VEGF在大肠癌的发生、发展过程中起重要作用。Objective To investigate the expression and clinical significance of phosphatidylinositol 3 kinase （PI3K）and vascular endothelial growth factor （VEGF） in the progression of colorectal cancer. Methods The protein expression and mRNA level were detected in 72 pairs colorectal carcinoma and the corresponding control tissue samples using real time polymerase chain reaction and immunohistochemistry methods. Results There were statistical significance in the differences of positive rates of PI3Kand VEGF between colorectal carcinoma and normal colorectal tissue （x^2= 60.66, ）（2= 83.70 ,Pall〈 0.05 ）, and the expression of PI3K and VEGF in colorectal carcinoma tissue showed positive correlation （r= 0.872, P〈 0.05）. The expression levels of genes PIK3R1 and VEGFA in colorectal carcinoma tissue were obviously higher than that of in the normal colorectal tissue, and the differences all had statistical significance（t= -2.75, t= -2.61, Pall〈 0.05）. Conclusion PI3K and VEGF play an important role in the occurrence and development of colorectal cancer.
