摘  要:选用枇杷野生近缘种‘栎叶枇杷’和栽培种‘早钟6号’、‘香妃’.对其叶绿体基因组trnH—psbA基因间隔区进行PCR克隆与测序.利用生物信息学方法分析其序列特征.并构建苹果亚科内19个属的系统发育树。结果表明:trnH-psbA序列长323~392bp(GenBank登录号:KF022254、KF022255、KF022256),G+C含量30.10%,33.13%,共有1个核苷酸替换(SUbstitution)和2个插入/缺失(indel);基于trnH—psbADNA条形码序列的分子系统进化分析结果表明.苹果亚科内形成2个大的分支,枇杷属与石斑木属的亲缘关系最近(平均遗传距离为0.055)。此结果表明叶绿体基因trnH-psbA序列可有效地进行枇杷属内物种鉴别及属间分类,是枇杷属植物DNA条形码研究的标准基因之一。The chloroplast genome trnH-psbA intergenic regions of three loquat (wild relatives 'E.prinoides', cultivars 'Zaozhong No.6' and 'Xiangfei' )were cloned and sequenced. The sequence characteristics were analyzed using bioinformatics methods and the phylogeny evolution tree was constructed with 19 genus of Maloideae. The results showed that the length of trnH-psbA sequence was 323-392 bp(GenBank accession number: KF022254, KF022255, KF022256), and its G+C content was 30.10%~33.13%. A total of 1 substitution and 2 indels were obtained in the data. Molecular phylogenetic anaylised based on the sequence of trnH-psbA DNA barcode reflected the relationship between Eriobotrya and Rhaphiolepis was closer(the average genetic distance was 0.055), and Maloideae was divided into two large branches. This paper suggested that trnH-psbA DNA barcode could be used as one of the standard gene to identify or classify plants of the Eriobotrya and its relative genus.
