摘 要:目的 :探讨乳房恶性肿瘤组织中端粒酶活性的表达情况及p16抑癌基因的影响。方法 :应用端粒重复放大程序 酶标法 (TRAP ELISA)及TRAP 银染法检测 35例乳癌组织及 2 4例良性肿瘤组织中的端粒酶活性 ,并采用多重PCR技术对乳癌组织的p16基因的缺失突变进行了分析。结果 :35例乳癌标本中有 2 8例端粒酶活性表达为阳性 (80 % ) ,而 2 4例乳房良性肿瘤标本中仅 1例端粒酶活性表达阳性 (4 1% )。此外 35例乳癌标本中的 15例存在有 p16基因的外显子 2及外显子 1的缺失 (4 2 8% ) ,而该 15例中有 14例被检测为端粒酶活性阳性 (93.3% ) ,但 2 0例未缺失标本的端粒酶阳性率仅为 70 % (14/2 0 )。结论 :端粒酶活性与乳房肿瘤组织的恶性程度密切相关 ,提示端粒酶参与了肿瘤的形成过程 ,而端粒酶活性的增高与p16抑癌基因的缺失突变之间可能存在有一定的关系。Objectives:To study the expression of telomerase activity and effect of deletion mutation of p16 gene in human breast cancer.Methods:Telomerase activity was detected in 35 cases of breast cancer and 24 cases of benign breast tumor by TRAP-ELISA and TRAP-silver staining.Using multiplex PCR,deletion mutations of p16 gene in the breast cancer were analyzed too.Results:Telomerase activity was detected in 28 of 35(80%) cases of breast cancer,but only detected in 1 of 24(4.1%) cases of benign tumor.Deletion mutation of the exon 2 and exon 1 of p16 gene was discovered in 15 of 35 cancer specimens(43%).14 of the 15 deletion mutational cases (93%) were showed telomerase activity,while the positive rate of telomerase activity was found only 70%(14/20) in the unmutational cases. Conclusions:Telomerase activity may play an important role in the development of breast cancer,and there may be a link between telomerase activity and deletion mutation of p16 gene.
