青蒿素介导肝癌细胞凋亡的实验研究

摘　　要：目的:探讨抗疟药青蒿琥酯对肝癌细胞株HepG_2的凋亡诱导作用.方法:用MTT测定、流式细胞术、梯状DNA电泳和透射电镜术检测细胞凋亡。结果:经青蒿琥酯处理的HepG_2细胞可见梯状DNA和凋亡小体等典型细胞凋亡特征.当青蒿琥酯浓度(80μmol/L)接近TC_(50)(74.21μmol/L)时,HepG_2细胞破坏率可达51.76%,细胞凋亡率为19.91%。结论:青蒿琥酯可诱导HepG_2细胞凋亡。Aim: To investigate apoptosis-induced effect of an antimalarial drug artesunate in hepatoma cell line HepG2. Methods : The apoptosis was detected by MTT assay, flow cytometry, ladder DNA electrophoresis and transmission electromicroscopy. Results: The apoptotic phenomena of the ladder DNAs and apoptotic bodies could be observed in the HepG2 cells treated with artesunate. When the concetration of artesunate (80pmol/L) was closed to TC50 (74. 21μmol/L), the breakdown rate and apoptosis rate of HepG2 cells were 51.76% and 19. 31%. Conclusions : Artesunate can induce apoptosis of HepG2.
