摘 要:目的:研究重组骆驼蓬脂转移蛋白(Peganum harmala lipid transfer protein,rPhLTP)对鼠黑色素瘤(B16)的体内抑制作用。方法:取B16细胞接种于C57BL/6J小鼠皮下,随机分为5组。测量各组瘤体积并称瘤重,计算抑瘤率。光镜观察脾脏、肝脏等变化;采用免疫组织化学法检测各肿瘤组织中血管内皮生长因子(VEGF)的表达。结果:高、中、低剂量处理组肿瘤相对体积(V/Vo)分别为3.21±0.12、3.57±0.07和3.92±0.42,抑瘤率分别为71.7%、58.1%和29.5%。显微镜下可见rPhLTP实验组肿瘤组织的细胞有不同程度的点、片状坏死。而肝、肺等无明显病理损伤。高剂量组中VEGF阳性表达指数低于生理盐水组(P<0.01),rPhLTP能明显抑制肿瘤血管的生成。结论:rPhLTP能有效地抑制小鼠B16细胞实体瘤的生长,其抗肿瘤的机制可能是通过抑制血管的生成来抑制肿瘤的生长。Objective:To evaluate the antitumor effect of recombinant Peganum harmala lipid transfer protein (rPhLTP) on the growth of murine melanoma(B16) ceils transplanted in mice. Method:B16 cells were inoculated subcutaneously of C57BL/6J mice,which randomly divided into 5 groups. Tumor volume and weight were measured, and the tumor inhibition rates were calculated. Meanwhile, we observed the change of the mouse spleen,liver,et al, under microscope. The expressions of vascular endothelial growth factor (VEGF) in tumor tissues were detected by immunohistochemistry. Result:rPhLTP inhibited the tumor growth in C57BIJ6J mice, with the relative tumor volume( V/ Vo) of 3.21 ± 0. 12,3.57 ± 0.07 and 3.92 ±0.42 and a tumor inhibition rate of 71.7% ,58.1% and 29.5% corresponding to rPhLTP dosage of 25,12. 5,6. 25 mg/kg, respectively. The treated tumor tissue cells have different levels of necrosis. The liver, lungs and other organs in mice haven' t significant pathological damage. The positive expression indexes of VEGF were significantly lower in high dose rPhLTP treatment groups than in NS group ( P 〈 0. 01 ). Conclusion : rPhLTP can inhibit the growth of solid tumor in C57BIM6J mice, suggesting that the antitumor effect of rPhLTP may be due to inhibition of tumor angiogenesis.
