生长素在人肝星状细胞中的抗纤维化作用

摘　　要：目的观察生长素(Ghrelin)对血小板源生长因子-BB(PDGF-BB)刺激的体外培养人肝星状细胞(HSCLX2)前胶原I合成及α-平滑肌肌动蛋白(α-SMA)生成的影响。方法体外培养HSC-LX2,分别行Ghrelin、PDGF-BB干预和按0.05、0.1、0.15μmol/L Ghrelin+PDGF干预细胞。采用定量PCR方法检测各组前胶原I mRNA的表达及Western印迹方法检测α-SMA表达情况。结果定量PCR结果显示PDGF处理组前胶原I mRNA的表达(6.91±0.46)较空白对照组(1.00±0.08)明显升高(P<0.05),证明PDGF能够促进HSC胶原合成。Ghrelin组前胶原I mRNA的表达(0.60±0.13)与空白对照组相比无明显变化,差异无统计学意义(P>0.05)。0.05、0.1、0.15μmol/L Ghrelin+PDGF共同处理组前胶原I mRNA表达分别为:(3.11±0.28)、(2.03±0.23)、(0.70±0.06),与PDGF组相比显著降低,差异有统计学意义(P<0.05),且随着Ghrelin浓度的增加其抑制作用越明显,呈浓度依赖性关系。Western印迹结果显示Ghrelin+PDGF共同处理组α-SMA较PDGF组表达降低。结论 PDGF刺激HSC活化后,Ghrelin可抑制其前胶原I及α-SMA生成,具有抗肝纤维化作用,可能成为防治肝纤维化的新途径之一。Objective To observe the effects of Ghrelin on synthesis of procollagen type I and expression of alpha- SMA from human hepatic stellate cells（HSC-LX2） stimulated by platelet-derived growth factor-BB（PDGF-BB）in vitro. Methods HSC-LX2 was cultured in vitro,and was treated by Ghrelin,PDGF-BB,or both of them according to different concentrations of Ghrelin,respectively.Procollagen I mRNA expression was detected by polymerase chain reaction（PCR）,andα-SMA expression was detected by western blot analysis in each groups.Results Expression of procollagen I mRNA in PDGF group（6.91±0.46） was significantly higher than that in blank control group（1.00±0.08）（P＆lt; 0.05） by PCR,which demonstrated that PDGF could promote synthesis of HSC procollagen type I.There was no significant difference in procollagen I mRNA expression between Ghrelin group （0.60±0.13） and blank control group（P＆gt;0.05）.Procollagen I mRNA expression,which was treated with different concentrations of both Ghrelin and PDGF（3.11±0.28、2.03±0.23 and 0.70±0.06）,respectively,was significantly reduced when compared to PDGF alone group（P＆lt;0.05）.The inhibition of Ghrelin was more significant with its concentration increaseα-SMA expression, which was treated with inboth Ghrelin and PDGF,was lower than that inwith PDGF alone by western blot.Conclusion After stimulated of PDGF in Hepatic stellate cell,Ghrelin could inhibit the synthesis of procollagen type I and alpha-SMA in the process of hepatic fibrosis.Thus,Ghrelin may become one of the new ways of in prevention prevention and treatmenment of liver fibrosis.
