川贝母对大鼠油酸型急性呼吸窘迫综合征的影响

摘  要:目的:观察川贝母对油酸所致急性呼吸窘迫综合征(acute respiratory distress syndrome,ARDS)大鼠肺组织的影响。方法:通过静脉注射油酸法建立大鼠ARDS模型。将40只模型大鼠随机均分为ARDS模型组,川贝母低剂量组,川贝母高剂量组,地塞米松组。另取10只Wistar大鼠作为空白对照组。在造模后0.5 h首次给药,1次/d,连续3 d。川贝母高、低剂量组分别给予5.0,2.5 mg/kg川贝母生药灌胃,空白对照组与模型组给予生理盐水灌胃,1 mL/100 g,地塞米松组给予地塞米松灌胃,10 mg/kg。测定各组大鼠左肺湿/干重比值(W/D),检测肺组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量;酶联免疫吸附法测定血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)的含量。结果:与空白对照组相比,模型组大鼠左肺W/D、MDA、TNF-α、IL-1β、IL-6含量显著升高,SOD活性降低(P均<0.05);与模型组相比,川贝母高、低剂量组及地塞米松组大鼠左肺W/D、MDA、TNF-α、IL-1β、IL-6含量显著降低,SOD活性升高(P均<0.05)。结论:川贝母可减轻油酸所致ARDS大鼠的肺组织病理损害程度,其机制可能是通过阻止体内脂质过氧化过程而实现。Objective: To observe the effect of tendrilleaf fritillary bulb on acute respiratory distress syndrome(ARDS) rats induced by oleic acid.Methods: The ARDS model was constructed by injection of oleic acid intravenously.Forty model rats were randomly divided into ARDS model group,low-dose tendrilleaf fritillary bulb group,high-dose tendrilleaf fritillary bulb group and dexamethasone group.Another ten Wistar rats were selected as the control group.Left lung wet / dry weight(W / D) ratio,lung tissue superoxide dismutase(SOD) activity,malondialdehyde(MDA) content were detected,serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6) levels were assayed by enzyme linked immunosorbent assay.Results: Compared with the control group,the model group showed lower levels of lung wet / dry weight(W / D) ratio,MDA,TNF-α,IL-1β,IL-6,but with higher levels of SOD activity(P 0.01 or P 0.05).The tendrilleaf fritillary bulb groups showed lower levels of lung wet / dry weight(W/D) ratio,MDA,TNF-α,IL-1β,IL-6,but higher levels of SOD,compared with the model group(both P 0.05).Conclusion: Tendrilleaf fritillary bulb could reduce lung tissue damage induced by oleic acid,and has the therapeutic effect on ARDS rat,which may be by preventing lipid peroxidation in the course of ARDS.
