摘 要:目的探讨高糖是否可以激活人肝星状细胞(hepatic stellate cells,HSCs)及其机制。方法实验分为正常对照组(NG组)、高糖组(HG组)和渗透压对照组(OC组);体外培养的人HSCs高糖处理后,应用westernblot方法检测TGF-β1、α-SMA、I型胶原和MMP-2的蛋白表达量;酶联免疫吸附试验(ELISA)测定TGF-β1的分泌水平。结果应用30μmol/L D-葡萄糖处理人HSCs细胞48h,可以激活HSCs,表现为:α-SMA、I型胶原和MMP-2的蛋白表达增加,TGF-β1的分泌水平升高。结论高糖可以激活人HSCs,进而促进肝纤维化的发生,此激活过程可能是通过TGF-β1通路。Objective To evaluate effects of high glucose on hepatic stellate cells and the mechanism. Methods The nomal control group(group NG),high glucose group(group HG) and osmotic pressure control group(group OC) were divided in this experiment. The expression of a-SMA(alpha-smooth muscle actin) ,collagen I and MMP-2 (matrix metalloproteinase 2)was measured by western-blot analysis. The secretion levels of TGF-β1 (transforming growth faetor-β1)were tested by enzyme-linked immunosorbent assay(ELISA). Results Treatment of cells with 30 μmol/L D-glucose significantly activated HSCs,as evidenced by increasing in secretion levels of TGF-β1 and protein expression levels of α-SMA,eollagen I,MMP-2. Conclusion High glucose activates HSCs and promoted the fibrosis process. These pathological processes might be mediated via the TGF-131 pathway.
