摘  要:目的 :为研究卵巢癌的免疫治疗方法构建合适的动物模型。方法 :严重联合免疫缺陷 (severecombinedimmunodeficiency ,SCID)小鼠皮下接种人卵巢癌细胞系SKOV3,同时腹腔注射人脾或外周血淋巴细胞进行免疫功能重建。观察成瘤情况 ;并用ELISA法检测鼠血清中人IgG含量以及流式细胞学和ABC免疫组织化学法测定鼠脾内人T细胞表型鉴定免疫重建。结果 :免疫重建的小鼠成瘤率为 89.6 % ,免疫组化证实皮下瘤保持原细胞系特征。 3种方法检测发现用人外周血淋巴细胞免疫重建的效果优于用脾淋巴细胞。结论 :免疫重建的荷人卵巢癌Objective: To establish an adequate experimental animal model for the development of immunotherapeutic strategies of ovarian carcinoma. Methods: Human ovarian cancer cell line SKOV 3 was administrated subcutaneously in severe combined immunodeficiency (SCID) mice, which was reconstituted by human lymphocytes from spleen or peripheral blood. The tumor growth was observed. Human immunoglobulins in mouse serum were assayed by ELISA, and human T cells in murine spleen were evaluated by flow cytometry and immunohistologic stainings respectively. Results: The rate of the reconstituted mice was 89.6%. Immunohistologic stainings showed that the subcutaneous tumors still had the characteristics of ovarian adenocarcinoma. The results illustrated that peripheral blood lymphocytes were better than spleen cells in the reconstitution. Conclusion:The reconstituted SCID mouse model with human ovarian cancer cell line has been established successfully.
