摘  要:目的考察泥鳅分泌多糖特征官能团含量及其体外抗氧化能力。方法采用水浸、醇沉、除蛋白、透析等方法提取泥鳅分泌多糖;采用乙酰丙酮分光光度法、硫酸咔唑法、硫酸钡比浊法依次检测泥鳅分泌多糖中氨基糖、糖醛酸、硫酸基含量;采用H2O2/Fe2+体系和邻苯三酚自氧化体系检测泥鳅分泌多糖体外清除羟基自由基和超氧阴离子自由基的能力。结果得到了纯度较好的泥鳅分泌多糖,其中氨基己糖和硫酸基含量较高,分别达到了11.19%和19.11%,而糖醛酸在样品中未检出。泥鳅分泌多糖具有清除羟基自由基和超氧阴离子自由基的能力,清除超氧阴离子自由基能力尤其明显。结论泥鳅分泌多糖中含有的氨基糖和硫酸基特征结构及其较为明显的清除氧自由基能力对其生物活性和药用价值具有重要意义。Objective To investigate characteristic functional groups content and antioxidative activity of polysaccharide from secretion of Misgurnus anguillicaudatus.Methods Polysaccharide from secretion of Misgurnus anguillicaudatus (MAP) was isolated and purified by lixiviation,ethanol precipitation,deproteinization and dialysis.Amino sugar content,uronic acid content and sulfuric acid group content of MAP were determined by the way of acetylacetone spectrophotometric method,sulfate-carbazole method and barium sulfate turbidimetric method,respectively.Scavenging ability of MAP to OH(-) and O(2-) were studied in the system of H2O2/ Fe2+ and pyrogallol auto-oxidation.Results MAP with ideal purification was obtained and relatively high contents of amino sugar (11.19%) and sulfuric acid group (19.11%) were found.There was no uronic acid existing in MAP under experimental condition.MAP showed scavenging ability on OH(-) and O(2-),the antioxidative effect on O(2-) was relatively obvious.Conclusion It is meaningful that MAP with characteristic functional groups and antioxidant effect displays bioactivity and medicine value.
