摘  要:目的:探讨二烯丙基二硫化物(DADS)对食管癌细胞株ECA-109体外侵袭能力的影响及其作用机制。方法:MTT法检测DADS对食管癌ECA-109细胞活力的影响,Transwell侵袭试验测定DADS在不同浓度时对ECA-109细胞侵袭能力的抑制作用。Western blot检测DADS在不同浓度下对ECA-109细胞MMP-2、MMP-9,TIMP-1、TIMP-2蛋白表达水平的影响。结果:DADS作用24h后,当浓度大于30μg/ml时,DADS可以显著抑制ECA-109细胞的活力,且随浓度升高其抑制作用相应增加;在浓度为10μg/ml-30μg/ml时,DADS能够明显减少穿过侵袭小室基底膜的肿瘤细胞数量,且抑制作用呈剂量依赖性;DADS可以使ECA-109细胞中MMP-2、MMP-9的表达水平降低,同时TIMP-1、TIMP-2的水平升高。结论:DADS在体外能够显著降低食管癌细胞的侵袭能力,其作用机制可能与DADS抑制MMP-2、MMP-9表达,上调TIMP-1、TIMP-2的表达有关。Objective:To investigate the effect and mechanism of diallyl disulfide(DADS) on invasion ability of esophageal cancer cells in vitro.Methods:MTT assay was executed to determine the inhibitory effect of DADS on ECA-109 cells,transwell chamber assay was used to determine DADS-induced effect on invasion ability of esophageal cancer cells.Western blot was used to detect DADS-induced effect on expression of MMP-2,MMP-9,TIMP-1,TIMP-2 protein in esophageal cancer cells.Results:DADS illustrate an inhibitory effect on ECA-109 cells over the concentration of 30μg/ml,DADS could significantly reduce the number of esophageal cancer cells through the invasion chamber in a dose-dependent manner.The expression levels of MMP-2,MMP-9 in esophageal cancer cell were reduced and at the same time the expression of TIMP-1,TIMP- 2 levels were significantly enhanced by DADS.Conclusion:DADS possess an ability of reducing invasion capacity of esophageal cancer cells,inhibiting the expression levels of MMP-2,MMP-9,and enhancing the levels of TIMP-1,TIMP-2.
