摘  要:目的研究甘遂毒性致大鼠血浆代谢组的变化,探讨代谢组学在中药毒性研究中的可行性。方法选取SD大鼠18只,适应1周后按随机数字表法分为3组:对照组、甘遂低剂量组(7.875 g/kg)和甘遂高剂量组(15.75 g/kg),持续给予甘遂2周后,取各组大鼠血浆,收集各组血浆样品的1H NMR谱,运用正交最小二乘判别(orthogonal partial leastsquares-discriminant analysis,OPLS-DA)方法分析。结果与对照组相比,甘遂组血浆代谢有明显变化。甘遂低剂量组血浆中乳酸和肌酸的含量升高(P<0.05),脂、3-羟丁酸盐、牛磺酸和葡萄糖的含量降低(P<0.05)。此外,丙氨酸、O-乙酰糖蛋白和氧化三甲胺在高剂量甘遂组中升高(P<0.05),而不饱和脂肪酸则降低(P<0.05)。结论甘遂可引起机体能量代谢、氨基酸代谢和脂代谢紊乱。Objective To clarify the plasma metabonomic changes induced by Kansui treatment revealing the toxicity mechanism of Kansui. Methods Eighteen SD rats were divided into 3 groups,i. e.,control,low dosage of Kansui group( 7. 875 g / kg) and high dose group( 15. 75 g / kg). After 2 weeks,the plasma samples were obtained,and 1H NMR spectra was performed and analyzed by orthogonal partial least-squares-discriminant analysis( OPLS-DA). Results There were some important changes in the plasma metabonomic profiles between Kansui treatment groups and control group. Plasma levels of lactate and creatine were increased( P〈0. 05),and those of lipids,3-hydrobutyrate,taurine and glucose were decreased in the 2 Kansui-treated groups( P〈0. 05). Meanwhile,the levels of alanine,O-acetyl-protein and trimethylamine-N-oxide were also elevated( P〈0. 05),and that of unsaturated fatty acid was decreased in high dosage Kansui treatment group( P〈0. 05). Conclusion Kansui induces the metabolic disorders of energy metabolism,amino acid and lipid metabolism.
