摘 要:目的探讨红益胶囊对-10°头低位卧床28 d诱导的猕猴下肢骨骼肌萎缩的保护作用,开发红益胶囊在肌萎缩防护中的新用途。方法采用-10°头低位卧床制备猕猴模拟失重性肌萎缩模型;猕猴分为对照组和喂食红益胶囊组,分别在卧床前(0天)和卧床后28 d取一侧比目鱼肌,应用免疫组织化学和实时定量PCR等方法,分析红益胶囊对肌萎缩形成的影响。结果 1)卧床28 d后,猕猴小腿平均围径和比目鱼肌肌纤维平均直径较卧床前显著下降(P<0.05),直径较小肌纤维含量明显增多,提示-10°头低位卧床28 d明显诱导了猕猴下肢骨骼肌和肌纤维的萎缩。2)卧床28 d后,猕猴比目鱼肌肌纤维横截面积明显下降,但红益胶囊组下降幅度较对照组显著减少(P<0.01)。3)-10°头低位卧床28 d后,肌萎缩特异因子Atrogin-1 mRNA的表达显著增加,红益胶囊组Atrogin-1 mRNA的表达受到了明显抑制(P<0.05)。结论红益胶囊可通过抑制肌萎缩特异因子Atrogin-1 mRNA的表达对抗-10°头低位卧床诱导的肌萎缩。提示了红益胶囊在肌萎缩防护中的新功能。Objective To explore the effects of Hongyi induced by - 10° head down bed rest(HDBR) for 28 capsules on rhesus monkey limb skeletal muscle atrophy d, and develop new usage of Hougyi capsules in protec- ting against muscle atrophy. Methods Rhesus monkey were divided into control and Hongyi capsules-treated group and fixed to bed in -10°HDBR to prepare simulated weightlessness induced muscular atrophy model. One side of the soleus muscle were removed at 0 and the 2gth day of - 10° HDBR respectively, then detected the effect of Hongyi capsules on muscle atrophy by immunohistochemistry and real time PCR. Results 1 ) After 28 d of - 10° HDBR, the average calf circumference and diameter of soleus fibers were decreased significantly ( P 〈 O. 05 ), and the distribution of muscle fibers showed significant increase of smaller fibers. It was demon- strated that -10° HDBR for 28 days significantly induced skeletal muscle fiber atrophy. 2)After 28 days of - 10 °HDBR, the cross-sectional area(CSA) of soleus muscle fiber from both groups were decreased, but the decrease was inhibited significantly in Hongyi capsules-treated group( P 〈 0.01 ) , which indicated that Hongyi capsules could effectively inhibit the decline of muscle fiber cross-sectional area during - 10° HDBR-induced muscle atrophy. 3) After 28 days of - 10° HDBR, Atrogin-1 mRNA expression was increased in both groups, but was notably inhibited in Hongyi capsules-treated group compared with control group ( P 〈 0. 05 ). Conclu- sion Hongyi capsules can resist muscular atrophy by inhibiting the expression of Atrogin-1 mRNA expression during -10° HDBR-induced muscle atrophy, which provides the new function of Hongyi capsules in protec- ting against muscle atrophy.
