肾透明细胞癌白细胞介素1β转换酶表达的研究

摘  要:目的 :研究肾透明细胞白细胞介素 1β转移酶 (interleukinlβ converting enzyme,ICE)的表达情况 ,探讨它在肾细胞癌 (RCC)细胞凋亡中的作用机制。方法 :采用免疫组织化学方法检测 41例肾透明细胞癌组织和 2 1例癌旁正常肾组织的 ICE表达 ,对 3例阳性细胞含量大于 10 %的肾癌组织进行 ICE和 TU NEL复染。结果 :ICE均为细胞质表达。正常肾组织阳性细胞含量为 5 9.8% ,肾透明细胞癌组织阳性细胞含量为 10 .4% ,二者差异有显著性 (P<0 .0 0 1) ;肿瘤组织的 ICE表达随病理分级的增高而降低。 ICE和 TUNEL复染显示二者不具有一致性。结论 :ICE在 RCC细胞凋亡早期发挥作用 ,与 RCC的发生。Objective:To study the ICE expression,and evaluate its mechanisms in the apoptosis of RCC.Methods:Immnohistochemistry and TUNEL(TdT mediated dUTP biotin nick end labeling)were used on 41 cases of RCC and 21 cases of normal tissue near the tumors mentioned above RCC section to detect ICE,double staining for ICE and TUNEL was done in 3 cases of RCC whose positive cell rate was higher than 10%.Results:ICE protein was confined to the cytoplasm.The positive cell rate in RCC(59.8%) was significantly lower than that in normal tissue(10.4%) (P<0.01 ).The expression of ICE showed an inverse relationship with nuclear grade of tumors.Double staining for ICE and TUNEL showed that it was as same as that of DNA fragmentation.Conclusion:ICE expression preceded DNA fragmentation during the apoptotic process,ICE may play an important role in RCC's development.
