摘 要:目的:探讨窖蛋白-1(Cav-1)在人肺浸润性腺癌(PIA)组织中的表达以及与自噬标志物——微管相关蛋白轻链3B(MAP LC3B)的关系及它们的临床意义。方法:利用量子点免疫荧光组织化学(QDs-IHC)技术检测Cav-1和LC3B蛋白在肺腺癌组织芯片包括68例PIA组织和10例非癌性肺组织中的表达;同时在10例PIA和10例非癌性肺组织中利用定量实时PCR检测Cav-1和LC3B mRNA的变化。结果:与非癌变肺组织相比,Cav-1和LC3B蛋白和mRNA在肺浸润性腺癌肿瘤细胞的表达均显著降低(P<0.05)。Cav-1在肿瘤细胞和基质的表达均与腺泡为主型PIA的病理分级显著相关(P<0.05),而与其他临床病理参数均无关(P>0.05)。LC3B在贴壁型的PIA阳性率为60.0%(9/15)显著高于腺泡为主型PIA的阳性率26.3%(10/38)(P<0.05),与其他临床病理参数均无关(P>0.05)。Cav-1和LC3B蛋白在肿瘤细胞的表达呈显著正相关(P=0.028,r=0.267)。结论:无论是在肿瘤细胞还是基质,Cav-1表达缺失可促进肺浸润性腺癌的形成,其机制可能与肿瘤细胞自噬不足有关。Objective: To detect the expression of caveolin marker-microtubule associated protein light chain 3 carcinoma (PIA) tissues, and to investigate their -l(Cav-1) and its correlation with autophagy (LC3B) in human pulmonary invasive adeno- clinicopathologic significance. Methods: The quantum dots immunofluorescent histochemistry (QDs-IHC) was used to detect the expression of Cav 1 and LC3B proteins in 68 cases of PIA and 10 cases of noncancerous lung tissues, and mR- NA levels of Cav-1 and LC3B were detected by quantitative real-time PCR (q-PCR) methods in 10 cases of PIA and 10 cases of noncancerous lung tissues. Results. lung tissues, proteins and mRNA levels of Car-1 and LC3B in the Compared with noncancerous PIA tissues were significantly decreased (P〈0.05). Expression of Cav-1 protein in the tumor cells and stroma was significantly related with histopathological grade of acinar predominant PIA (P〈0. 05), but not correlated with the other clinicopathologic parameters (P〉0.05). Positive rates of LC3B protein in the lep- idic predominant PIA was 60.0% (9/15), significantly higher than 35. 71% (10/28) of acinar predominant PIA (P〈0. 05). There was positive correlation between Car-1 and LC3B in the tumor cells (P〈0.05, rs =0. 267). Conclusion. Absent expression of Cav-1 in the tumor cell and stroma may promote pulmonary invasive adenocarcinoma formation and its mechanism may corre- late with inadequate autophagy in the tumor cells.
