摘 要:目的探讨信号转导与转录激活因子1(STAT1)、干扰素-γ(IFN-γ)在口腔扁平苔藓(OLP)病损发生发展中的作用。方法 48例OLP及10例正常口腔黏膜石蜡包埋组织,采用免疫组化SABC法检测相应组织中STAT1和IFN-γ蛋白的表达情况,分析其与OLP患者临床病理特征的相关性。结果①STAT1、IFN-γ在OLP病损区表达阳性率均高于正常对照组,差异有统计学意义(P<0.05)。②OLP病损区STAT1阳性表达率在IFN-γ阳性组高于IFN-γ阴性组,差异有统计学意义(χ2=5.042,P<0.05);IFN-γ和STAT1表达之间存在正相关关系(r=0.324,P<0.05)。③STAT1阳性表达率在病程短于6个月OLP组高于病程长于6个月OLP组,差异有统计学意义(χ2=5.077,P<0.05);IFN-γ阳性表达与基底细胞液化程度呈负相关(r=-0.388,P<0.05)。结论 OLP局部组织中存在STAT1和IFN-γ蛋白高表达,IFN-γ/STAT1通路在OLP局部病损形成过程中起到推动作用。Objective To understand the possible role of signal transducer and activator of transcription 1 (STAT1) and interferon-γ (IFN-γ) in the generation of local lesions of oral lichen planus (OLP). Methods Formalin-fixed, paraffin-embedded tissues of 10 cases of normal oral epithelium and 48 cases of OLP were investigated for STAT1 and IFN-γ proteins by using immunohistochemistry SABC method. The correlation between the expression of STAT1 and IFN-γ proteins and the clinical and pathological characteristics in OLP were statistically analyzed. Results ① STAT1 and IFN-γ were over expressed compared with healthy oral mueosa (P〈 0.05). ② The percentage of STAT1 over-expression in OLP with positive IFN-γ staining was significantly higher than that in OLP with negative IFN-γ staining (x^2 =5. 042, P〈0.05). A positive correlation between the over- expression of STAT1 and IFN-γ was seen in OLP (r=0. 324, P〈0.05). ③The percentage of over-expression of STAT1 in OLP with disease course less than 6 months was higher than that in OLP with disease course more than 6 months (x^2 =5. 077, P〈0. 05), and a positive correlation was observed between the over-expression of IFN-γ and the liquefaelion of base cells in local lesions of OLP (r= -0. 388, P〈0. 053. Conclusion The over-expression of STAT1 and IFN-γ may participate in the generation of local pathological damnification of OLP at the early stage.
