摘 要:目的观察静脉输注全氟化碳(PFC)预处理对气管内滴注脂多糖(LPS)致急性肺损伤(Au)大鼠的保护作用及相关机制。方法42只雄性Wistar大鼠随机分为七组,每组6只,即对照组(NS组),LPS2、4、6h组,PFC2、4、6h组。LPS组、PFC组分别经股静脉注射生理盐水6mL/kg、PFC6mL/kg,30min后气管内滴注LPS1mg/kg,NS组以等容量生理盐水作对照。Ns组于气管内滴注生理盐水后6h,PFC组与LPS组分别于滴注LPS后2、4、6h处死动物。比较各组PaO2、肺湿干比(W/D)、肺病理学变化、肺髓过氧化物酶(MPO)及细胞间黏附分子-1(ICAM-1)、血中性粒细胞CDllb表达情况。结果与LPS组比较,PFC组大鼠W/D、MPO活性、CDllb及ICAM-1表达水平均下降(P〈0.05),PaO2升高,肺组织损伤减轻。结论静脉输注PFC乳剂对LPS致ALI具有保护作用,机制可能与下调ICAM-1及CDllb表达有关。Objective To investigate the protective effect and the possible mechanism of intravenous infusion with perfluorocarbon (PFC) on acute lung injury (ALI) induced by intratracheal instillation of lipopolysaccharide (LPS) in rats. Methods 42 Wistar rats were randomly divided into 7 group(n = 6) : control group ( NS group), LPS 2, 4, 6 h group and PFC 2, 4, 6 h group. Normal saline was injected into LPS group via the femoral vein 6 mL/kg 30 min before intratracheal instillation of LPS 1 mg/kg. PFC was injected into PFC group via the femoral vein 6 mL/kg 30 min before intratracheal instillation of LPS 1 mg/kg. Normal saline was given to NS group as a control. NS group were sacrificed 6 h after being injected with NS. LPS group and PFC group were sacrificed 2, 4, 6 h after being poisoned. PaO2, wet to dry weight ratio of lung (W/D), pathological changes of lung, expression of myeloperoxidase (MPO) and intercellular adhesion molecule- 1 (ICAM - 1 ) of lung and CDllb in peripheral blood neutrophils (PMN) were assessed. Results Compared with LPS group, W/D, MPO, CDllb and ICAM - 1 were significantly decreased PaO2 was increased and lung injury was alleviated in PFC group (P 〈 0.05). Conclusion Intravenous infusion of PFC emulsion significantly protects lung from LPS- induced acute lung injury in rats probably by the suppression of ICAM -1 and CD1 l b.
