摘　　要：目的:研究及探讨蛋白酶体抑制剂硼替佐米(Bortezomib,PS-341)联合放疗对卵巢癌SKOV3/DDP细胞株生长及凋亡的影响。方法:采用四甲基偶氮唑蓝(MTT)法检测不同放射剂量的单药PS-341、DDP及两药联合对SKOV3/DDP细胞株生长的抑制率,并分析PS-341的耐药逆转作用及其放射敏感性。结果:①不同浓度的PS-341、DDP及不同剂量的放疗可以抑制卵巢癌SKOV3/DDP细胞株的体外生长,且随着PS-341浓度的增加及作用时间的延长,能有效地抑制细胞株的增殖,两药联合可增强DDP对卵巢癌SKOV3/DDP细胞株体外生长的抑制作用。②卵巢癌SKOV3/DDP细胞株对DDP及放疗具有交叉抗性,PS-341联合DDP能增强SKOV3/DDP细胞株对体外放疗的敏感性。结论:①PS-341能有效抑制SKOV3/DDP细胞株的增殖,从而逆转其对DDP的耐药性,促进卵巢癌SKOV3/DDP细胞株的凋亡,与DDP具有协同作用,且时间和剂量呈依赖关系;②PS-341联合DDP对SKOV3/DDP细胞株具有放疗增敏作用,有效抑制细胞株的增殖。Objective： To explore the combined effect of proteasome inhibitior bortezomib and radiotherapy on the growth of the ovarian cancer SKOV3/DDP cell lines. Methods： Different radiation dose of PS - 341, DDP and PS - 341 ＋ DDP combined were given to explore the inhibition rate of SKOV3/DDP cell growth, the resistance reuersal effect of PS- 341 and the radiosensitivity of the ovarian cancer SKOV3/DDP cell lines by MTT. Results： Different concentrations of PS -341, DDP and different doses of radiation could suppress the vitro growth of ovarian cancer SKOV3/DDP cells, besides, with the increase of concentration of PS - 341 and the time of implication which could effectively inhibit the proliferation of cell limes, the combination of the both drugs could enhance the inhibition of DDP growth of ovarian cancer SKOV3/DDP. Ovarian cancer SKOV3/DDP cells had cross - resistance to DDP and radiotherapy, the combination of PS - 341 and DDP could enhance the vitro radiation sensitivity of SKOV3/DDP cells. Conclusion： First, Bortezomib can effectively inhibit the proliferation of SKOV3/DDP cells, and reverses its resistance to DDP, thus promote the apoptosis of SKOV3/DDP cells. It demonstrates a combined effect with DDP, besides, it is depended with time and dose closely. Secondly, the combination of PS -341 and DDP on SKOV3/DDP has radiosensitizer action, which can inhibit the proliferation of cells effectively.
