摘  要:目的在体外探讨6-环己基甲氧基鸟嘌呤(O6-Cyclohexylmethylguanine,NU2058)对结直肠癌细胞生长及增殖的影响。方法分别设NU2058组、5-氟尿嘧啶(5-fluorouracil,5-FU)组、NU2058与5-FU联用组,通过MTT法检测其对HCT-8细胞生长的抑制作用,应用流式细胞术检测其对HCT-8细胞周期及细胞凋亡率的影响。结果 NU2058对HCT-8细胞生长有明显的抑制作用,24 h IC50值为12.01 mg/L,48 h IC50值为22.93 mg/L,两者有明显的差异。联合5-FU作用48 h后抑制效果增强,但两种药物之间无明显相互促进作用。NU2058对细胞周期有明显影响,表现为G0/G1期细胞减少,S期细胞增多。结论 NU2058对HCT-8细胞的生长具有明显的抑制作用,且与时间及浓度变化成正相关;其对肿瘤增殖的抑制作用与细胞周期阻滞途径相关。[Objective] To investigate the influence of NU2058 on the growth and proliferation of col orectal cancer ceils in vitro. [Method] HCT-8 cells were divided into three groups. They were NU2058 group, 5-FU group and NU2058 combined with 5-FU group, respectively. The proliferation inhibition rate was detected by MTT assay, and the apoptosis ratio and cell cycle blockage were examined by flow cytometry. [Result] NU2058 had inhibitive effect on the growth of HCT-8, which improved when combined with 5-FU. But these two drugs did not interact with each other. Half maximal inhibitory concentration (IC50) of NU2058 required to treat HCT-8 cells for 24 h was percentage of cells in Go/Gl phase reduced significantly while in creased in S period relatively. [Conclusion] NU2058 has apparently inhibitive effect on the growth of HCT- 8, and its drug effect is 12.01 rag/L, while for 48 h was 22.93 mg/L. NU2058 affected cell cycle apparently. The positively related to time and dose; its tumor proliferation inhibitive effect is relevant to cell cycle in hibitive pathway.
