摘  要:探究油茶果皮多酚粗提物对DNA氧化损伤的保护作用。采用单细胞凝胶电泳技术(SCGE)和琼脂糖凝胶电泳技术(AGE),分别研究不同浓度的油茶果皮多酚粗提物对H2O2诱导人脐静脉内皮细胞(HUVEC)DNA损伤的保护作用,以及对APPH损伤质粒pGL6DNA的保护作用。经SCGE分析显示:H202诱导细胞DNA严重损伤,在经50~1000mg/L药物作用后,阴性对照组的细胞拖尾率和彗星尾长与阳性对照组相比均有显著的下降(P〈0.01或P〈0.05);而经1500mg/L的药物预处理后,细胞拖尾率和彗星尾长与阳性对照组相比无显著性差异(P〉0.05);经AGE分析,油茶果皮多酚粗提物对APPH介导的DNA链断裂有显著的保护作用。油茶果皮多酚粗提物能有效地清除羟自由基,从而起到保护DNA的作用。To investigate the effect of Camellia pericarp polyphenol crude extracts on DNA damage. Using single cell gel electrophoresis and agarose gel electrophoresis, different concentrations of Camellia pericarp polyphenol crude extracts were investigated its protective effect on DNA damage of HUVEC induced by H2O2 ,as well as plas- mid pGL6 DNA induced by APPH. Result The analyzed results of SCGE showed that the cells induced by H2O2 was damaged serious- ly. Under the conditions of 50, 100, 500, 1 000 nag/L, the comet rate and comet tail length in negative control group were significantly lower than those in positive control(P〈0.01), while after pretreat- ment by 1 500 mg/L Camellia pericarp polyphenol crude extracts. The comet rate and comet tail length had no significant difference compared with positive control group(P〈0.05). Analysis by agar- ose gel eleetrophoresis, Camellia pericarp polyphenols crude extracthad a significant protective effect on DNA strand breaks induced by APPH. Camellia pericarp polyphenol crude extracts could effectively scavenge hydroxyl free radicals, thus protecting the role of DNA.
