摘  要:目的:探讨小细胞肺癌c-myc癌基因的表达对Nrf2表达的影响,分析c-myc与Nrf2表达的相关性。方法:选用两株耐药性不同的肺小细胞癌肿瘤细胞株,小细胞肺癌药物敏感株H69及耐药株H69AR,Western-Blot方法比较Nrf2,c-myc在两株细胞间的表达差异;H69细胞通过Lipofectamine2000脂质体转染过表达c-myc,Western-Blot方法检测过表达c-myc前后Nrf2蛋白表达变化;H69AR细胞通过Lipofectamine2000脂质体转染c-myc siRNA,Western-Blot方法检测c-myc siRNA前后Nrf2的蛋白表达变化。结果:与H69细胞相比,H69AR细胞株中c-myc及Nrf2高表达;H69细胞c-myc转染后,Nrf2表达上调;H69AR细胞c-myc siRNA干扰后,Nrf2表达下调。结论:小细胞肺癌中Nrf2的表达与c-myc表达呈正相关。Objective:To determine the influence of oncogene c-myc over Nrf2 in small cell lung cancer, and to an- alyze the correlation between c-myc and Nrf2 expressions. Methods: Two lines of small-cell lung cancer cells were chosen in this study. One is drug sensitive H69 cell. The other is multidrug resistant H69AR cell. Western blot assays were uti- lized to compare the different expression levels of c-myc and Nrf2. Overexpression of c-myc by Lipofectamine 2000 trans- fection was performed in H69 cells, and Nrf2 protein expression was compared between the control and c-myc transfection groups. Down-regulation of c-myc by siRNA transfection was performed in H69AR cells, and Nrf2 protein expression was compared between the control and e-myc siRNA groups. Results: Compared with H69 cells, protein expression level of c- myc and Nrf2 was much higher in H69AR cell. In H69 tumor cells, Nrf2 was up-regulated after c-myc transfection. Mean- while, in H69AR tumor cells, Nrf2 was down-regulated after c-mye siRNA transfection. Conclusion: Nrf2 and c-myc is positively correlated in small cell lung cancer. To determine the influence of c-myc over Nrf2 in small cell lung cancer, and to analyse the correlation between c-myc and Nrf2 expressions.
