摘  要:目的研究tempol氮氧自由基的酚类衍生物对辐射细胞及小鼠损伤的保护作用。方法合成了4种兼具tempol氮氧自由基和酚羟基的化合物,比较它们对肿瘤细胞和正常细胞的辐射防护作用,并进一步测定被辐照小鼠外周血的白细胞数(WBC)、中性粒细胞数(ANC)、单核细胞数(AMC)、血小板数(platelet count)等指标。结果合成并表征了4个tempol酚类衍生物结构。与tempol相比,辐射小鼠的WBC,ANC,AMC,血小板数具有不同程度的改善。结论稳定的氮氧自由基是一类较好的抗氧化损伤药物,tempol的酚类衍生物在细胞和动物水平均显示了较好的辐射防护能力。Obj ective To investigate the bioactivity of tempol-phenol conj ugates on radiation-induced damage in cells and irradiated mice.Methods Four compounds with both tempol and phenol substructure were synthesized.Their protective effect on normal cells compared to tumor cells was evaluated.Their effect on peripheral WBC,ANC,AMC and platelet counts in an rat model invi-vo was determined.Results The four conjugated derivatives were prepared and characterized.Invivo study showed that compared to the tempol group,the compounds treated group had greater recovery in WBC,ANC,AMC and platelet count.Conclusion Stable nitroxides are the potential antioxidant drugs.The Tempol-phenol conj ugates have an improved radiation protect activity both in cells and mice.And further research on the mechanism should be taken into consideration.
