摘  要:目的:探讨健脑益智方对脑缺血再灌注损伤后白细胞计数、髓过氧化物酶及细胞间黏附分子-1的影响。方法:采用健康雄性SD大鼠四动脉闭塞法制备大鼠全脑缺血再灌注模型,于全脑缺血20 min,再灌注24 h后,测定外周血白细胞总数、非淋巴细胞数及百分比、淋巴细胞数及百分比;大脑皮质和海马中MPO的活性及海马结构内ICAM-1表达情况。结果:模型组大鼠外周静脉血中的白细胞总数、非淋巴细胞计数及其百分比明显升高(P<0.01),淋巴细胞百分比降低(P<0.01);大鼠脑组织皮质区和海马区的MPO含量均明显升高(P<0.01,P<0.05);ICAM-1阳性神经元平均光密度值高于假手术组(P<0.01),平均灰度值低于假手术组(P<0.01)。与模型组比较,阿司匹林干预后造模组、中药干预后造模组及中药治疗组的白细胞总数、非淋巴细胞总数(P<0.01)及其百分比均降低(P<0.05),淋巴细胞百分比升高(P<0.05);MPO的含量均不同程度的降低(P<0.01,P<0.05);ICAM-1阳性神经元平均光密度值均降低(P<0.01,P<0.05),平均灰度值均升高(P<0.01,P<0.05)。结论:健脑益智方可抑制大鼠脑缺血再灌注后缺血脑组织中的白细胞(主要是中性粒细胞)的浸润,下调脑组织中细胞间黏附分子-1的表达,并可能通过以上途径阻断其后的炎症级联反应来达到减轻脑缺血再灌注损伤,促进神经功能的修复。Objective:To study the effects of Jiannao Yizhi Recipe on white blood cell count, myeloperoxidase and intercellular adhesion molecule-1 induced by cerebral ischemia-reperfusion injury in rats. Method: Four-vessel occlusion cerebral ischemia-reperfuion was selected to establish the model of male SD healthy rats. After the rats were global cerebral ischemic for 20 minutes, reperfusion for 24 hours, white blood cell, nonlymphoid leukocytes lymphocytes and the percentage of them were assayed; myeloperoxidase activity in cortex and hippocampus was tested; the expression of intercellular adhesion m01ecule-1 in hippocampus was reflected. Result:Total number of peripheral venous white blood cell, the number of non-lymphocyte and non-lymphocytic percentage of model group were elavated ( P〈0.01 ). The lymphocytic percentage was decreased ( P〈0.05 ); the content of myeloperoxidase in cortex and hippocampus of model group was significantly elevated ( P〈0.01, P〈0.05 ); the average optical density of intercellular adhesion molecule-1 in the positive neurons was higher than that of sham operation ( P〈0.01 ). The while the average gray value was lower (P〈0.01). Compared with model group, in Chinese herbs intervention group, aspirin intervention group and Chinese herbs treatment group, the total numbers of white blood cell and non-lymphocytic percentage were decreased (P〈0.05), while lymphoc, yte percentage was elevated (P〈0.05); content of myeloperoxidase appeared different degrees decrease;the average optical density of intercellular adhesion molecule-1 in the positive neurons was decreased ( P〈0.01, P〈0.05 ), and the average gray value of intercellular adhesion molecule-1 in the positive neurons was increased ( P〈0.01, P〈0.05 ). Conclusions : Jiannao Yizhi Recipe can inhibit leukocyte infiltration ( such as neutrophil ) in ischemic brain tissue, down regulate the expression of intercellular adhesion molecule-1, which may depend on these ways mentioned abov
