摘  要:为研究杜仲α-亚麻酸的合成机制,用BLAST序列比对工具对杜仲果实和叶片的转录组数据进行分析,在转录组中获得了13条α-亚麻酸合成相关酶基因,其中成熟果实和幼果差异表达基因6条,叶片和幼果中的差异表达基因3条;酰基ACP硫脂酶(EuFatA)基因在幼果和成熟果实表达量无显著差异;硬脂酰-ACP去饱和酶(SAD)基因至少由4个家族成员(EuSAD1~EuSAD4)组成,成熟果实中4个家族成员均有表达,而幼果中只有EuSAD1表达,幼果和成熟果实中的表达量均以EuSAD1为主;δ-12脂肪酸脱氢酶(FAD2)基因家族至少由5个家族成员组成,EuFAD2-1和EuFAD2-2在幼果和成熟果实中表达量的差异达到显著水平,EuFAD2-1-EuFAD2-3的表达量果实均高于叶片,而EuFAD2-4和EuFAD2-5幼果高于成熟果实,调控机理较复杂;ω-3脂肪酸脱氢酶(FAD3)基因至少由6个成员组成,仅有EuFAD3-6在成熟果实和幼果中的表达量有显著差异。In order to study the synthesis mechanism of α-linolenic acid, the transcriptome data in fruits and leaves of Eucommia ulmoides were analyzed by BLAST sequence alignment tool. 13 related enzyme genes are obtained, in which there are six differential expression genes in young fruit and mature fruit, and three differential expression genes in leaf and young fruit. The gene expressive levels of EuFatA gene in young fruit and mature fruit show no significant difference. SAD gene family includes four members at least (EuSADI-EuSAD4), all of the four members express in mature fruit, only EuSAD1 expresses in young fruit, and EuSAD1 mainly express in young fruit and mature fruit. FAD2 gene family includes five members at least, and their regulatory mechanism is complex. The expression of EuFAD2-1 and EuFAD2-2 in young fruit and mature fruit reach the significant level, the expression levels of EuFAD2-1 and EuFAD2-3 in fruit are higher than those in leaf, while the expression levels of EuFAD2-4 and EuFAD2-5 in young fruit are higher than those in mature fruit. FAD3 gene family includes six members at least, and only the expression of EuFAD3-6 in young fruit and mature fruit has significant differences.
