隔离子两侧临近基因的共表达研究

摘  要:染色质隔离子是相邻转录功能区的边界序列,可以发挥增强子阻断和染色质屏障作用.研究结果显示,隔离子两侧临近基因的转录方向相同时,这两个基因表达水平具有显著的正相关,否则表达水平关联不显著.该结果指出隔离子并没有对临近基因表达起到屏障作用,这些隔离子可能发挥了增强子阻断作用.此外,隔离子两侧转录方向相同的两个基因的基因长度呈现正关联.尤其是同时位于"+"链的隔离子两侧临近的基因,关联最强.同时,发现隔离子每一侧基因表达水平和该基因长度呈现显著的负相关,这一关联性与该基因的转录方向和隔离子的存在无关.Chromatin insulators are border sequences in neighboring functional regions of tran-scription, which can play a role in enhancer blocking and chromatin barrier. The results show a posi-tive correlation between expression levels of two neighboring genes flanking insulators when the transcriptional directions of the two genes are the same. Otherwise,there is not a signifieant correla- tion. These results suggested insulators could not exert effects of chromatin barrier for expressions of neighboring genes. The insulators might play a role in enhancer blocking. Furthermore,there is a positive correlation between gene lengths of two neighboring genes flanking insulators when tran- scriptional directions of the two genes are the same. Especially there is a very strong association be-tween the two genes located on "+" strands simultaneously. Meanwhile it is found that gene ex-pression levels are significantly and negatively correlated with gene lengths at eaeh side of insula- ors. The correlations are not related to transcription directions of genes and existence of insulators.
