摘 要:目的探讨兔高脂饮食建立动脉硬化模型后wnt1、β-catenin及dvl-1的变化及高脂饮食引起动脉粥样硬化的可能机制。方法选择3-4月龄健康雄性日本大耳白兔16只,随机分为正常对照组、粥样硬化组,通过高脂饮食建立兔动脉硬化模型,12周后HE染色显微镜下观察主动脉病理形态改变,用免疫组化和Western blot法检测主动脉壁wnt1、β-catenin及dvl-1的表达。结果正常对照组主动脉三层结构清晰,内膜光滑完整,平滑肌细胞排列整齐;粥样硬化组内膜明显增厚,可见大小不等、形态不规则的泡沫细胞聚集,弹力板处及细胞间隙可见大量脂质沉积,平滑肌细胞排列不规则;粥样硬化组中wnt1、β-catenin及dvl-1的表达水平较正常对照组显著增加(P<0.05)。结论高脂饮食可引起动脉粥样硬化,可能与激活Wnt/β-catenin信号通路有关。Objective The aim of this study was to investigate the variation in the expression of wntl, β-catenin, and dvl-1 in a rabbit arteriosclerosis model established by high fat diet, and to evaluate the underlying mechanism of atherosclerosis induced by high fat diet. Methods Sixteen 3--4 months old healthy Japanese white male rabbits were selected and randomly divided into the normal control group and the atherosclerosis model group. Atherosclerosis model was established by high fat diet and subjected to examination of the aorta pathological feature under the HE staining microscopy at the end of this 12-week study. Immunohisto- chemistry and western blot were used to monitor the expression of wntl, β-catenin, and dvl-1 on the aorta wall. Results For the normal control group, the three layer structure of the aorta was distinct; the intima was smooth and complete; the smooth muscle cells were arranged normally. For the atherosclerosis model group, the intima was markedly thicken; aggregation of foam cells in different size and morphology were noticed; a large number of lipid deposition were observed in elastic lamina and intercellular space; and the smooth muscle cells were arranged irregularly. The expression levels of wntl, β -catenin, and dvl-1 of the atherosclerosis model group were significantly higher than those of the normal control group (P〈O.05). Conclusion High fat diet may cause atherosclerosis and the mechanism may involve activation of Wnt/β -catenin signaling pathway.
