摘 要:目的探讨人白血病骨髓间充质干细胞(LMSC)对K562细胞生长曲线及细胞周期分布的影响。方法采用噻唑蓝(MTT)法,绘制与LMSC共培养前后K562细胞的生长曲线;流式细胞仪检测与LMSC共培养前后,K562细胞周期的变化。结果与LMSC共培养后,K562细胞生长受抑,G0~G1期细胞比例明显增高(47.3±1.3)%,S期细胞比例明显下降(39.1±0.6)%,二者与K562细胞单独悬浮培养组[G0~G1期:(39.6±1.2)%,S期:(48.0±1.2)%]相比,差异均有统计学意义(P〈0.05);与LMSC共培养后,G2~M期细胞比例虽然也呈上升趋势,但差异无统计学意义(P〉0.05)。结论①与LMSC共培养后K562细胞生长受抑制。②LM—SC通过使K562细胞周期阻滞于G0~G1期发挥抑制K562细胞生长的作用。Objective To compare the proliferation and cell cycle progression of K562 cells that cultivated with or without LMSC. Methods K562 cells cultivated in suspension and adhesively cultivated with MSCs were collected respectively and cell proliferation curves were drawn. The cell cycle of K562 cells after co-cultured with LMSC was detected by flow cytometry. Results After cultured with LMSC, the proliferation capacity of K562 cells were inhibited. Cell cycle progression was suppressed when co-cultured K562 cells with LMSC. Cell cycle distribution suggested that the proportion of cells in G0-G1 stage increased while that in S stage decreased after the exposure of K562 cells to LMSC (P〈0.05). Although the proportion of cells in G2-M stage presented a trend to rise, there was no distinct variety (P〈0.05). Conclusion LMSC can suppress cell cycle progression of K562 cells in G0-G1 stage, thus inhibit the proliferation of K562 cells.
