摘 要:目的:探讨牙周慢性炎症对牙周膜干细胞(periodontal ligament stem cells,PDLSCs)生物学特性的影响。方法:体外有限稀释法克隆化培养健康个体和慢性牙周炎患者牙周膜干细胞(H-PDLSCs和P-PDLSCs),免疫荧光染色法分析细胞表面分子;克隆形成和Edu染色检测细胞增殖能力;并通过体外成骨、成脂、成软骨诱导培养,比较两组PDLSCs多向分化能力。结果:慢性牙周炎患者牙周膜组织中可分离培养出PDLSCs,与分离自牙周健康者的H-PDLSCs相比,其增殖能力升高,但分化能力减弱(P<0.05)。结论:牙周慢性炎症微环境可降低PDLSCs的分化潜能。AIM: To investigate the effects of chronic periodotatal inflammatory microenvironment on the biological characteristics of periodontal ligament stem cells (PDLSCs). METHODS:PDLSCs were isolated by the limited dilution technique from the periodontal ligament tissue of healthy individuals and the patients with periodontitits respectively (H-PDLSCs and P-PDLSCs). The proliferation and clone formation of the cells were studied by Edu staining and clone formation assay, the osteogenic, adipogenic and chondrogenic differentiation was determined by multi - directional differentiation assay. RESULTS : PDLSCs were successfully obtained from the periodontal ligament tissue of both healthy individuals and the patients with periodontitis. P-PDLSCs exhibited decreased muhipotent differentiation potential and increased proliferation ability compared with H-PDLSCs (P 〈 0.05). CONCLUSION: The chronic inflammatory microenvironment may inhibite the differentiation potential of PDLSCs.
