摘 要:目的:研究双酶水解鹿茸胶原蛋白制备鹿茸胶原多肽。方法:以梅花鹿鹿茸为原料,采用热水提取鹿茸胶原蛋白,先后加入2种酶酶解,测定酶解后鹿茸胶原多肽相对分子质量分布,根据单因素试验和正交试验确定两种酶的用量和酶解时间。结果:鹿茸胶原多肽的最佳工艺条件为:胃蛋白酶与鹿茸比(g:g)为1:100,酶解12 h,胰蛋白酶与鹿茸比(g:g)为1:100,酶解1 h,酶解得到的鹿茸胶原多肽相对分子质量小于3000约占60.65%,相对分子质量小于5000的约占88.16%,提取率为11.16%,蛋白质含量为89.07%。结论:采用双酶法提取制备鹿茸胶原多肽,相对分子质量小,易被人体消化吸收,该研究为鹿茸胶原多肽产品的开发和应用奠定了基础。Objective: this essay aims at studying the preparation of antler collagen polypeptide through hydrolyzing antler collagen by double enzyme. Method: Antler collagen was firstly extracted by hot water from sika deer antler which was employed as the raw material; then after the enzymolysis of the two kinds of enzymes added, the distribution of molecular weight of antler collagen polypeptide was measured, and the enzyme concentration and the time of enzymolysis were obtained through single factor experiment and orthogonal test. Results: The optimal technological condition for obtaining antler collagen polypeptide is as follows. The proportion of pepsin and antler is 1:100(g:g) and the time for enzymolysis is 12 h, and the proportion of trypsin and antler is 1:100(g:g) and the time for enzymolysis is 1 h. Antler collagen polypeptides obtained from enzymolysis whose molecular weight are below 3000 account for approx. 60.65%, those below 5000 account for approx. 88.16%. The extraction rate is 11.16%, the content of protein is 89.07%. Conclusion: Antler collagen polypeptide obtained by way of double enzyme method has the features of lower molecular weight and easily being absorbed by human body. This study will lay the foundation for further development and application of antler collagen polypeptide product.
