摘  要:目的:以野葛中主要异黄酮的含量为指标探讨河南野葛质量评价方法。方法:采用HPLC法测定不同产地野葛中葛根素大豆苷、大豆苷元的含量,ODSHYPERSIL色谱柱(4.6 mm×250 mm,5μm);乙腈-水梯度洗脱;柱温30℃;流速0.8 mL/min;检测波长250 nm。结果:葛根素、大豆苷、大豆苷元分别在0.1632~1.3056、0.0700~0.5632、0.004796~0.03837μg内线性关系良好;平均回收率分别为101.51%,98.08%,101.04%;相对标准偏差(RSD)分别为1.11%、1.35%、2.12%;结论:本实验建立的方法简便、准确、重复性好,可以用于河南不同产地野葛的质量评价。Objective: To evaluate the quality of radix puerariae lobatae in different origin of Henan province based on the contents of main isoflavones in it. Methods: Establish a method of HPLC for quantitative determination of puerarin, daidzin and daidzein in radix pu- erariae lobatae. The procedure of HPLC was performed on the chromatographic column of ODS HYPERSIL(4. 6 mm × 250 mm,5 μm), and the mobile phase in the gradient elution program was acetonitrile-water. The column temperature was 30 ℃, the flow rate was 0. 8 mL/min, and the detection wavelength was 250 nm. Results:The three compounds showed good linear relationship within the range of 0. 1632 - 1. 3056,0. 0700 - 0. 5632 and 0. 004796 - 0. 03837 μg. The average recovery were 101.51% ( RSD = 1.11% ), 98.08% (RSD = 1.35% ) , 101.04% (RSD = 2. 12% ) respectively. Conclusion: The method is easy and accurate with higher repeatability, which can be used in the quality control of radix puerariae lobatae.
