THBS4基因在高原地区胃癌组织中表达的生物学行为及临床意义

摘　　要：目的通过研究高原地区胃癌组织中THBS4基因的表达,探讨THBS4基因对高原地区胃癌生物学行为的影响及为高原地区胃预后的评估提供理论依据。方法收集高原地区无化疗且临床资料完整的胃癌手术标本60例,采用RT-PCR及S-P法,检测60例胃癌标本中THBS4蛋白的表达。结果用RT-PCR技术分析的结果显示,10例高分化胃癌组织中THBS4 mRNA有5例表达;29例低分化胃癌中24例表达,高分化癌与中、低分化癌组织中的表达有差异。中、低分化胃癌中的THBS4 mRNA水平高于高分化胃癌(P<0.05)。THBS4mRNA表达与淋巴转移有关,但与临床分期无关。免疫组化S-P法染色分析显示,THBS4蛋白在低分化29例中21例表达;中分化阳性率8例;均高于高分化(4例),THBS4在高分化癌和中、低分化癌组织中的表达差异有统计学意义(P<0.05);中、低分化胃癌中的的THBS4阳性高于高分化胃癌。THBS4基因在高原地区胃癌组织中表达与胃癌的分化程度有关;与淋巴转移有关。结论 THBS4基因可作为判断高原地区胃癌细胞分化不良的指标,对胃癌的预后评估有重要意义。Objective To investigate the biological behaviors and clinical significance of expression of t hrombospondin4 （THBS4） in gastric cancer tissues at plateau region in order to provide theoretical basis for prognosis evaluation of gastric cancer in plateau area. Methods The expression levels of THBS4 protein in 60 gastric cancer specimens in plateau area were detected by RT-PCR and S-P. Results RT-PCR results showed that there were 5 cases that expressed THBS4 mRNA among 10 cases of well differentiated gastric cancers and there were 24 cases that expressed THBS4 mRNA among 29 cases of poorly differentiated gastric cancers, there was an obvious difference in the expression of THBS4 mRNA between well differentiated gastric cancers and moderately, poorly differentiated gastric cancers. The expression levels of THBS4 mRNA in moderately, poorly differentiated gastric cancers were significantly higher than those in well differentiated gastric cancers （ P 〈 0.05）. The expression of THBS4mRNA was correlated to lymphatic metastasis, however, which was not related to clinical stage. Immunohistochemistry （SP） results showed that there were 21 cases that expressed THBS4 protein among 29 cases of poorly differentiated gastric cancers and there were 8 cases that expressed THBS4 protein in moderately differentiated gastric cancers,which were higher than that （4 cases） in well differentiated gastric cancers, and there was an significant difference in the expression of THBS4 between well differentiated gastric cancers and moderately, poorly differentiated gastric cancers （ P 〈 0.05 ）. The positive rates of THBS4 expression in moderately, poorly differentiated gastric cancers were high than those in well differentiated gastric cancers. The expression of THBS4 gene in plateau area was correlated to differentiation degree and lymphatic metastasis of gastric cancers.
