精神分裂症患者血清蛋白表达差异的研究

摘　　要：目的采用蛋白质组学技术筛选精神分裂症患者血清蛋白标志物,为该疾病寻找客观的临床诊断依据。方法利用CM10芯片和表面增强激光解吸电离飞行时间质谱(SELDI-TOF-MS)技术检测健康对照组和精神疾病组的血清蛋白质谱。将筛查出的特异蛋白质峰通过查询蛋白数据库查找出相对应的蛋白质。结果在健康对照组和精神疾病组中,筛选出差异有统计学意义的蛋白质峰15个(P<0.01),相对分子质量分别为2 058.06±0.70、2 159.68±0.60、2 651.54±0.42、2 774.36±0.35、2 912.61±1.69、3 116.9±0.81、3 168.95±1.82、3 407.71±1.33、4 050.38±0.68、4 100.61±0.36、5 642.83±1.69、5 913.05±0.16、9 186.42±3.01、2 093.81±1.46、2 221.92±1.82。结论精神分裂症患者与健康人相比存在有特异性的血清蛋白,这些特异的血清蛋白对该病的诊断具有一定价值。Objective To use the proteomics technique to screen the serum protein markers in the patients with schizophrenia for seeking the objective clinical diagnosis basis of this disease. Methods The SELDI-TOF MS technique and CM10 chip were adopted to detect serum protein spectrum in the mental diseases group and the healthy control group. Tile corresponding proteins for screened specific protein peaks were found by querying the protein da- tabase. Results hi the mental disease group and the healthy control group,15 protein peaks with statistical difference were screened out（P〈0.01）. Their relative molecular weights were 2 058.06±0.70,2 159.68±0.60,2 651.54± 0.42,2 774.36±0.35,2 912. 61±1. 69,3 116. 9±0. 81,3 168. 95±1. 82,3 407.71±1. 33,4 050. 38±0. 68, 4 100.61±0.36,5 642.83±1.69,5 913.05±0. 16,9 186.42±3.01,2 093.81±1.46 and 2 221.92±1.82 respec- tively. Conclusion Compared with normal subjects, schizophrenia patients have specific serum protein, which may have certain diagnostic value in the diagnosis of this disease.
