摘  要:目的:研究miR-125a-5p对卵巢癌SKOV3细胞增殖和侵袭的影响。方法:通过慢病毒转染技术将含miR-125a-5p的质粒转入SKOV3细胞中,经嘌呤霉素筛选建立稳定表达mi-125a-Sp的SKOV3细胞株。通过实时定量荧光RT-PCR验证miR-125a-5p的表达情况,MTT、流式技术、Transwell、划痕实验检测过表达miR-125a-5p后对SKOV3细胞增殖、凋亡、侵袭及转移能力的影响。结果:过表达miR-125a-5p后,SKOV3细胞增殖能力较对照组有显著提高;流式细胞检测显示过表达miR-125a-5p的SKOV3细胞S期细胞增多,GO/GI期细胞及细胞凋亡率减少;Transwell和划痕实验显示过表达miR-125a-5p的SKOV3细胞侵袭和迁移能力明显高于对照组。结论:SKOV3细胞miR-125a-Sp过表达可能通过减少G0/G1期细胞及细胞凋亡率从而促进细胞增殖、侵袭和迁移能力。Objective: To explore effects of miR-125a-5p on multiplication and invasion in SKOV3 of ovarian cancer ceils. Methods: Using lentivirus transfection technology, plasmids of miR-125a-5p were transformed into SKOV3 and selected by puromycin to establish:SKOV3 cell strains to express miR-125a-Sp steadily. Proving the expressing situation of rniR-125a-Sp by RT-PCR, the proliferation, cell cycle, apoptosis rate, invasion and migration of SKOV3 after the overexpression of miR- 125a-Sp were analysed by MTq', flow cytometric (FCM), transwell and scratch test. Results: The ability of proliferation of SKOV3 was improved obviously compared with the control group after overexpression of miR-125a-Sp; the S-phase of the cells after the overexpression of miR-125a-Sp was increased obviously, G0/G1 phase and the apoptosis rate were decreased; transwell and scratch test results indicated that the cells after the overexpression of miR-125a-Sp had a better invasion and migration ability than did the control group. Conclusion: SKOV3 after overexpression of miR-125a-Sp may promote itself to improve the ability of proliferation, invasion and migration by decreasing G0/G1-phase calls and the apoptosis rate of cells.
