摘  要:目的:通过观察丝胶对Ⅱ型糖尿病大鼠肾基质金属蛋白酶-9(MMP-9)/基质金属蛋白酶组织抑制因子-1(TIMP-1)的调节作用,探讨丝胶保护糖尿病。肾损伤的作用及机制。方法:链脲佐菌素腹腔注射建立Ⅱ型糖尿病大鼠模型,丝胶治疗组大鼠给予丝胶灌胃、阳性对照组大鼠给予二甲双胍灌胃。ELISA法检测各组大鼠血清Ⅳ型胶原和层黏连蛋白的含量;免疫印迹和RT-PCR法检测各组大鼠肾MMP-9和TIMP-1蛋白和mRNA的表达。结果:丝胶治疗组较糖尿病模型组,血清Ⅳ型胶原和层黏连蛋白的含量明显降低,肾MMP-9的表达明显升高,TIMP-1的表达明显降低;丝胶治疗组较阳性对照组无明显差别。结论:丝胶可通过恢复糖尿病时肾MMP-9/TIMP-1的平衡改善肾细胞外基质代谢,从而减轻肾小球硬化和肾间质纤维化,延缓糖尿病肾损伤的发生发展。Objective: To investigate the protective effects and mechanisms of sericin on diabetes kidney injury by observing the regulating effects of sericin on matrix metalloproteinase-9 (MMP-9)/tissue inhibitors of matrix metalloproteinases-1 (TIMP- 1) in diabetes mellitus rats. Methods: 48 adult healthy male SD rats were randomly divided into 4 groups with 12 rats in each group: a normal control group, a diabetes mellitus model group, a sericin treatment group and a positive control group. Type 2 diabetes mellitus model was made by injecting streptozotocin into the peritoneal cavity continuously, and then the rats were lavaged with sericin (2.4 g/kg) in the seriein treatment group and metformin (55.33 mg/kg) in the positive control group. The type IV collagen and laminin content in serum of rats were detected by ELISA. Western blotting and RT-PCR were used to detect the expression of MMP-9 and TIMP-1 in the kidneys. Results Compared with the rats in the diabetes mellitus model group, the type IV collagen and laminin content in serum of the rats in the sericin treatment group was decreased obviously; the expression of MMP-9 was up-regulated, TIMP-1 expression was down-regulated significantly in the sericin treatment group. The type IV collagen and laminin content in serum and the expression of MMP-9 and TIMP-1 in the sericin treatment group had no obvious differences compared with the positive control group. Conclusion: Sericin can improve extracellular matrix metabolism in the kidney by recovering MMP-9/TIMP-1 balance during diabetes mellitus, so as to lighten glomerulosclerosis and renal interstitial fibrosis and delay development of kidney injury.
