摘 要:目的:探讨高尔基体基质蛋白130(Golgi matrix protein 130,GM130)、14-3-3ζ和整合素β3在卵巢上皮恶性肿瘤组织中的表达及其临床意义。方法:应用免疫组织化学法检测49例卵巢上皮恶性肿瘤、30例卵巢上皮良性肿瘤和23例正常卵巢上皮组织中GM130、14-3-3ζ和整合素β3的表达,RT-PCR和蛋白质印迹法分别检测卵巢上皮恶性肿瘤、卵巢上皮良性肿瘤和正常卵巢上皮组织中GM130、14-3-3ζ和整合素β3 mRNA及蛋白的表达。结果:GM130、14-3-3ζ和整合素β3在卵巢上皮恶性肿瘤组织中的阳性表达率明显高于卵巢上皮良性肿瘤和正常卵巢上皮组织,差异有统计学意义(P<0.05)。卵巢上皮恶性肿瘤组织中GM130与14-3-3ζ的表达、14-3-3ζ与整合素β3的表达和GM130与整合素β3的表达均呈正相关(r=0.517,P<0.05;r=0.415,P<0.05;r=0.384,P<0.05)。GM130、14-3-3ζ和整合素β3的表达与卵巢上皮恶性肿瘤患者的临床分期和组织分化程度有关(P<0.05),与年龄、肿瘤大小和病理类型无关(P>0.05)。卵巢上皮恶性肿瘤组织中GM130、14-3-3ζ和整合素β3 mRNA及蛋白的表达水平明显高于卵巢上皮良性肿瘤和正常卵巢上皮组织(P<0.05)。结论:GM130、14-3-3ζ和整合素β3在卵巢上皮恶性肿瘤中的高表达可能在其恶性进程中发挥重要作用,GM130有望成为判断卵巢癌患者预后的指标及治疗靶点。Objective: To investigate the expressions of Golgi matrix protein 130 (GM130), 14-3-3ξ and integrinβ3 in ovarian epithelial malignant tumor tissues and their clinical significance. Methods: The expressions of GM130, 14-3-3ξ and integrinβ3 in 49 ovarian epithelial malignant tumor tissues, 30 ovarian epithelial benign tumor tissues and 23 normal ovarian epithelial tissues were detected by immunohistochemistry. The expression levels of GM130, 14-3-3ξ and integrin133 mRNAs and proteins in ovarian epithelial malignant tumor tissues, ovarian epithelial benign tumor tissues and normal ovarian epithelial tissues were detected by RT-PCR and Western blotting, respectively. Results: The positive rates of GM130, 14-3-3ξ and integrinβ3 expressions in ovarian epithelial malignant tumor tissues were higher than those in the ovarian epithelial benign tumor tissues and normal ovarian epithelial tissues (P 〈 0.05). There were positive correlations between the expressions of GM130 and 14-3-3ξ (r = 0.517, P 〈 0.05), 14-3-3ξ and integrin133 (r =- 0.415, P 〈 0.05), and GM130 and integrin133 (r = 0.384, P 〈 0.05) in ovarian epithelial malignant tumor tissues. The expressions of GM130, 14-3-3ξ and integrin133 were obviously associated with the clinical stage and pathological differentiation of ovarian epithelial malignant tumor (P 〈 0.05), but not be associated with age, tumor size and pathological type (P 〉 0.05). The expression levels of GM130, 14-3-34 and integrin133 mRNAs and proteins in ovarian epithelial malignant tumor tissues were higher than those in the ovarian epithelial benign tumor tissues and the normal ovarian epithelial tissues (P 〈 0.05). Conclusion: The high expressions of GM130, 14-3-34 and integrin^3 in ovarian epithelial malignant tumor may play an important role in malignant progression. GM130 may become a prognostic factor and a therapeutic target in patients with ovarian tumors.
