摘 要:目的:探讨缺氧诱导因子-1α(hypoxia-inducible factor,HIF-1α)和血管内皮生长因子(vascular endothelial growth factor,VEGF)在肝细胞癌(hepatocellular carcinoma,HCC)的表达及其与肿瘤血管生成的关系。方法:采用免疫组织化学SP法检测41例HCC组织及16例非肿瘤肝组织的HIF-1α及VEGF表达,采用鼠抗人CD31单克隆抗体标记肿瘤血管内皮细胞,计数HCC组织微血管密度(micro-vessel density,MVD)值。结果:在HCC组织与非肿瘤肝组织中,HIF-1α、VEGF阳性表达率和MVD值分别为82.9%、80.5%、41.0±20.2与37.5%、25.0%、20.1±9.1,两组差异均具有统计学意义(P<0.05)。在HCC组织中,HIF-1α、VEGF与MVD三者之间均具有相关性,并与病理分级相关(P<0.05)。结论:在HCC组织中,HIF-1α和VEGF呈高表达,与肿瘤血管生成密切相关。Objective:Explore and discuss the expression of hypoxia-inducible factor-1 α( HIF-1 α) and vascular endothelial growth factor (VEGF) in hepatocellular carcinoma as well as their correlations with tumor angiogenesis. Methods :The HIF-1α and VEGF ex- pression in Forty-one cases of HCC tissue samples and 16 cases of nontumorous hepatic tissue samples were analyzed by immunohisto-chemistry streptavidin-peroxidase (SP) method. The anti-human mouse CD31 monoclonal antibody was adopted to mark the tumor vas- cular endothelial cells and assess the micro-vessel density (MVD). Results:The positive expression rates of HIF-1α and VEGF were 82.9% (34/41),37.5% (6/16),80.5% (33/41) and 25.0% (4/16) and the MVD values were 41.0 ±20.2 and 20.1 ±9.1 in HCC tissues and adjacent noncancerous tissues respectively;the differences between the two groups were statistically significant for all three variables (P 〈 0. 05 ). The expression levels of HIF-1α ,VEGF and the MVD value are correlated with each other in HCC tissues. Moreover,HIF-1α and VEGF expression,as well as the MVD value,were correlated with the pathological degree of HCC. Conclusion: HIF-1 α and VEGF are frequently over expressed in HCC,and play an important role in tumor angiogenesis.
